猪源microRNAs靶向非洲猪瘟病毒基因区域分析

Analysis of swine-derived microRNAs targeting African swine fever virus gene region

摘要 为了探讨宿主microRNAs(miRNAs)是否靶向非洲猪瘟病毒(ASFV)基因,以及初步确定miRNAs靶向病毒基因的区域,试验以ASFV感染猪差异表达的miRNAs为研究对象,利用miRanda软件分析miRNAs靶向ASFV基因的具体区域和结合能力,再利用MEGA 7.0软件分析miRNAs所靶向的ASFV基因序列的保守性。结果表明:ssc-miR-122和ssc-miR-125b可同时靶向ASFV CP2475L基因,其中ssc-miR-122靶向该基因4个区域;ssc-miR-181a和ssc-miR-125b可同时靶向ASFV MGF_300-4L基因,其中ssc-miR-181a靶向该基因2个区域;此外,ssc-miR-181a可靶向ASFV NP1450L、E111R基因,ssc-miR-125b靶向ASFV P1192R基因,ssc-miR-145-5p靶向ASFV NP868R、C717R、MGF_505-1R等8个基因,ssc-miR-126-3p靶向ASFV G1211R、EP296R、MGF_110-5L-6L基因,ssc-miR-92a靶向ASFVI329L和EP424R基因;选择多个miRNAs所靶向的ASFV CP2475L和MGF_300-4L基因序列构建进化树,CP2475L和MGF_300-4L基因序列同源性均为100%。说明猪源miRNAs具有潜在的靶向抑制ASFV复制的能力,初步明确了ssc-miR-122、ssc-miR-181a、ssc-miR-125b等miRNAs靶向ASFV CP2475L、MGF_300-4L等基因的具体区域。 In order to investigate whether host microRNAs(miRNAs)target African swine fever virus(ASFV)genes,and to preliminarily determine the regions of viral genes targeted by miRNAs,in the experiment,the differentially expressed miRNAs of ASFV-infected pigs were used as the research objects.The specific regions and binding abilities of miRNAs targeting ASFV gene were analyzed by miRanda software,and the conservation of ASFV gene sequences targeted by miRNAs was analyzed by MEGA 7.0 software.The results showed that ssc-miR-122 and ssc-miR-125 b could simultaneously target the ASFV CP2475 L gene,and ssc-miR-122 targeted 4 regions of the gene.The ssc-miR-181 a and ssc-miR-125 b could simultaneously target the ASFV MGF_300-4 L gene,of which ssc-miR-181 a targeted two regions of the gene.The ssc-miR-181 a targeted ASFV NP1450 L,E111 R genes;ssc-miR-125 b targeted ASFV P1192 R gene;ssc-miR-145-5 p targeted 8 genes including ASFV NP868 R,C717 R,MGF_505-1 R;ssc-miR-126-3 p targeted ASFV G1211 R,EP296 R,MGF_110-5 L-6 L genes;ssc-miR-92 a targeted ASFV I329 L and EP424 R genes.The ASFV CP2475 L and MGF_300-4 L gene sequences targeted by multiple miRNAs were selected to construct a phylogenetic tree.The homology of CP2475 L and MGF_300-4 L gene sequences was 100%.The results suggested that porcine-derived miRNAs had the potential to target and inhibit the replication of ASFV,and preliminarily determined the specific regions of miRNAs such as ssc-miR-122,ssc-miR-181 a,ssc-miR-125 b targeting ASFV CP2475 L,MGF_300-4 L and other genes.
