摘要
目的采用超高效液相色谱-四级杆/飞行时间串联质谱(UPLC-Q-TOF-MS)法定性分析黑参灌胃给药后大鼠血清、尿液及粪便中的化学成分。方法采用Waters Acquity UPLC BEH C_(18)色谱柱(2.1 mm×100 mm,1.7μm),以乙腈(A)-0.1%甲酸水溶液(B)为流动相,梯度洗脱;流速:0.3 mL·min^(-1);柱温:30℃;进样量:10μL。电喷雾离子化源(ESI);检测模式为正(ESI^(+))/负(ESI^(-))离子检测模式,灌胃黑参提取物后,对大鼠血清、尿液及粪便中的成分进行定性分析。结果共鉴定出黑参提取物中17种人参皂苷成分,其中血清、尿液及粪便分别检出12种、8种、11种原型皂苷,并在粪便中检测到3种代谢产物。结论首次研究黑参中的人参皂苷类成分在大鼠体内的代谢情况,可为黑参的药效物质基础研究及其开发利用提供参考。
Objective To determine the chemical constituents from Black Ginseng Radix et Rhizoma in serum,feces and urine of rats by ultra-performance liquid chromatography-quadrupole/time of flight tandem mass spectrometry(UPLC-Q-TOF-MS). Methods A Waters Acquity UPLC BEH Ccolumn(2.1 mm×100 mm,1.7 μm)was used with acetonitrile(A)-0.1% formic acid aqueous solution(B)as mobile phase in gradient elution. The flow rate was 0.3 mL·minand column temperature was set at 30 ℃,and the injection volume was 10 μL. Electrospray ionization source(ESI)was used. The detection mode was positive(ESI)/negative(ESI-)ion detection mode. The components in serum, urine and feces of the rats were qualitatively analyzed after intragastric administration of Black Ginseng Radix et Rhizoma extract. Results A total of 17 ginsenosides were identified in the extract of Black Ginseng Radix et Rhizoma,among which 12,8 and 11 protosaponins were detected in serum,urine and feces,respectively, and 3 metabolites were detected in feces. Conclusion The metabolism of ginsenosides from Black Ginseng Radix et Rhizoma in rats was described for the first time,which can provide a reliable scientific evaluation for the basic research on the pharmacodynamic substances of Black Ginseng Radix et Rhizoma,and provide a basis for the further development and utilization of Black Ginseng.
