葛根素对肥胖小鼠血压和主动脉内皮功能的影响 被引量:9

摘要 目的探究葛根素(puerarin,Pue)对高脂饮食诱导的肥胖小鼠主动脉内皮功能和血压的影响。方法♂C57 BL/6J小鼠,随机分为5组,每组各6只:正常饮食组(Con)、高脂饮食组(DIO)、高脂饮食+低剂量葛根素组(20 mg·kg^(-1)·d^(-1))、高脂饮食+中剂量葛根素(40 mg·kg^(-1)·d^(-1))组、高脂饮食+高剂量葛根素组(80 mg·kg^(-1)·d^(-1)),记录体质量和血压。8周后测空腹血糖,检测血脂含量;检测主动脉血管舒张功能;分离原代内皮细胞进行Transwell和细胞增殖实验。结果①高剂量葛根素干预后,可减轻高脂饮食引起的肥胖小鼠体质量、体脂、血糖、血压的升高,与DIO组相比,明显降低,P<0.01;②高剂量葛根素干预可改善主动脉的舒张功能,与DIO相比,P<0.01;③葛根素可改善DIO小鼠原代内皮细胞的迁移能力,与DIO相比,P<0.01。结论葛根素可以明显降低饮食诱导的肥胖小鼠血压,其机制可能是改善主动脉内皮舒张功能和内皮细胞增殖迁移能力。 Aim To explore the effect of puerarin(Pue)on aortic function and blood pressure in hypertensive mice induced by high-fat diet.Methods Thirty male mice were divided into five groups named as normal diet group(Con),high-fat diet group(DIO),high-fat diet+low-dose puerarin group(20 mg·kg^(-1)·d^(-1)),high-fat diet+medium-dose puerarin(40 mg·kg^(-1)·d^(-1))group and high-fat diet+high-dose puerarin group(80 mg·kg^(-1)·d^(-1)).The mice were injected intraperitoneally with Pue for eight weeks.Body weight,blood pressure and blood glucose were measured.Serum was collected to detect blood lipid.Aortas were separated from aortic endothelial cells to test the vasodilative function.Aortic endothelial cells from DIO mice were isolated to perform Transwell and cell proliferation experiments.Results①High-dose puerarin treatment could reduce the body weight,body fat,blood glucose and blood pressure in obese mice(P<0.01);②High dose of puerarin could improve the vasodilative function of aortas compared with those from DIO mice(P<0.01);③The migration ability of primary endothelial cells from DIO+Pue group was improved compared with that from DIO group(P<0.01).Conclusions Puerarin can significantly reduce blood pressure in obese mice induced by high fat diet by improving the aortic diastolic function and endothelial cell proliferation and migration.
