培土生金法对慢性阻塞性肺疾病大鼠肺泡、膈肌及其线粒体病理形态的影响 被引量:1

Effects of Therapy of Reinforcing Earth to Strengthen Metal on the Pathomorphology of Pulmonary Alveoli,Diaphragm and Its Mitochondria in Rats with Chronic Obstructive Pulmonary Disease

摘要 【目的】探讨培土生金法对慢性阻塞性肺疾病(COPD)大鼠的治疗作用及机制。【方法】将48只大鼠随机分为空白组,模型组,阳性药物组和中药高、中、低剂量组,每组8只。除空白组之外,其余各组大鼠采用烟熏法建立COPD模型。造模第21天开始给药,中药高、中、低剂量组分别对应给予1.8、0.9、0.45 g(生药)/kg(体质量)培土生金法中药复方灌胃,阳性药物组给予地塞米松0.2 mg/kg灌胃,每天1次,直至第60天结束实验。给药结束后,分别对肺和膈肌进行病理检查。【结果】与空白组比较,模型组的肺泡和膈肌组织破坏明显,肺泡面积明显增大,线粒体和肌丝排列不规则,结构紊乱;与模型组比较,阳性药物组和中药各剂量组的肺泡和膈肌组织相对完整,肺泡面积相对减少,膈肌细胞中线粒体和肌丝排列规则,结构相对完好。【结论】培土生金法可改善COPD大鼠肺泡、膈肌及其线粒体损伤,能保护肺泡、膈肌及其线粒体结构的完整性,提示培土生金法具有提高COPD大鼠肺泡、膈肌及其线粒体功能,改善呼吸肌疲劳,减少呼吸衰竭发生的作用。 Objective To explore the therapeutic effects and mechanism of Therapy of Reinforcing Earth to Strengthen Metal(invigorating spleen to strengthen lung)on pulmonary alveoli,diaphragm and mitochondria in rats with chronic obstructive pulmonary disease(COPD).Methods The 48 rats were randomly divided into the blank group,the model group,the positive drug group and the Chinese herbal medicine high-,medium-and low-dose groups,8 rats in each group.Apart from the blank group,the COPD rat model was established by smoking in the other groups.On the 21st day of modeling,the Chinese herbal medicine high-,medium-and lowdose groups were given 1.8,0.9 and 0.45 g(raw drug)/kg(body mass)of Chinese herbal compound prescription with Therapy of Reinforcing Earth to Strengthen Metal respectively by gavage,while the positive drug group was given 0.2 mg/kg of dexamethasone by gavage,once a day,until the end of the experiment on the 60th day.At the end of the administration,the pathological examinations of the lungs and diaphragm were performed separately.Results Compared with the blank group,the pulmonary alveoli and diaphragm tissues of the model group were obviously destroyed,the pulmonary alveoli area was significantly increased,the mitochondria and myofilaments were irregularly arranged and structurally disordered;compared with the model group,the pulmonary alveoli and diaphragm tissues of the positive drug group and Chinese herbal medicine dose groups were relatively intact,the pulmonary alveoli area was relatively less,and the mitochondria and myofilaments in the diaphragm cells were regularly arranged and relatively structurally intact.Conclusion Therapy of Reinforcing Earth to Strengthen Metal has improved the damage of pulmonary alveoli,diaphragm and its mitochondria in COPD rats,and it has protected the structural integrity of pulmonary alveoli,diaphragm and its mitochondria,suggesting that Therapy of Reinforcing Earth to Strengthen Metal may has the effect of improving the function of pulmonary alveoli,diaphragm and their mitochondria,alleviating respiratory muscle fatigue and reducing the occurrence of respiratory failure in COPD rats.
