臻通集胶囊对高脂血症模型大鼠的治疗作用及其机制研究 被引量:2

Study on the Treatment and Mechanism of Zhentongji Capsule(臻通集胶囊)on Hyperlipidemia Model Rats

摘要 目的:探讨臻通集胶囊对高脂血症模型大鼠的疗效及其作用机制。方法:随机取12只SD大鼠作为空白组,正常饮食;取88只大鼠进行造模,给予高脂饲料喂养4周后,测定TG和CHO。取造模成功的72只大鼠根据CHO值随机分为模型组、西药组、中西药联合组、中药高剂量组、中药中剂量组、中药低剂量组。各药物组大鼠灌胃给予相应药物,空白组和模型组大鼠灌胃给予等体积的生理盐水,1次/d,连续给药8周后,麻醉采血,称体质量和肝脏质量,计算肝脏系数;检测血清中血脂水平TG、CHO、LDL、HDL和TBA的表达水平,并计算H/L值;检测载脂蛋白Apo-A1和Apo-B100的表达水平;检测MDA、SOD和T-AOC等氧化应激水平,同时检测肝脏组织中胆固醇合成相关酶HMG-COA和ACAT的表达;HE染色进行病理观察。结果:与空白组比较,模型组大鼠体质量及肝脏系数,血清中CHO、TG、LDL、HDL、Apo-A1、Apo-B100、MDA含量,肝脏组织中HMG-COA、ACAT含量均明显升高(P<0.05),而血清中SOD、T-AOC活性及H/L均明显降低(P<0.05);与模型组比较,西药组、中西药联合组和中药高剂量组大鼠体质量及肝脏系数均明显降低(P<0.05),中药中剂量组大鼠肝脏系数明显降低(P<0.05);与模型组比较,西药组、中西药联合组、中药高剂量组、中药中剂量组大鼠血清CHO、TG、LDL含量均明显降低(P<0.05),H/L均明显升高(P<0.05);中西药联合组、中药高剂量组大鼠血清HDL含量均明显低于模型组(P<0.05);与模型组比较,中西药联合组、中药高剂量组大鼠血清中Apo-A1、Apo-B100含量均明显降低(P<0.05),西药组和中药低剂量组Apo-B100显著降低(P<0.05);与模型组比较,中西药联合组、中药中剂量组、中药高剂量组大鼠血清中MDA含量明显降低(P<0.05),而SOD、T-AOC活性明显升高(P<0.05);与模型组比较,中西药联合组、中药高剂量组大鼠肝脏组织中HMG-COA、ACAT含量均明显降低(P<0.05);西药组大鼠肝脏组织中HMG-COA含量明显低于模型组(P<0.05),中药中剂量组大鼠肝脏组织中ACAT含量明显低于模型组(P<0.05)。结论:臻通集胶囊能够有效降低高脂血症大鼠的血脂水平,与其抗氧化、抑制胆固醇合成等密切相关。 Objective:To explore the curative effect of Zhentongji Capsule on hyperlipemia model rats and its preliminary mechanism.Methods:12 SD rats were randomly selected as the blank group with normal diet.88 rats were fed with high-fat diet for 4 weeks,and TG and CHO were measured.72 successful rats were divided into model group,western medicine group,combined Chinese and western medicine group,Chinese medicine high,middle and low dose group according to CHO value according to CHO value.Rats in each drug group were given corresponding drugs by gavage.Rats in the blank group and model group were given the same volume of normal saline by gavage,once a day.After 8 weeks of continuous administration,blood was collected under anesthesia,body mass and liver mass were weighed,and liver coefficient was calculated.The expression levels of lipid level TG,CHO,LDL,HDL and TBA in serum were detected,and the H/L value was calculated.The expression levels of apolipoprotein Apo-A1 and Apo-B100 were detected.The oxidative stress levels of MDA,SOD,T-AOC were detected,and the expression of cholesterol synthesis related enzymes HMG-COA and ACAT in liver tissue was detected.He staining was used for pathological observation.Results:Compared with the blank group,the body mass and liver coefficient,the contents of CHO,TG,LDL,HDL,Apo-A1,Apo-B100 and MDA in the serum,the contents of HMG-COA and ACAT in the liver tissue of the model group were significantly increased(P<0.05),while the activities of SOD,T-AOC and H/L in the serum were significantly decreased(P<0.05).Compared with the model group,the body mass and liver coefficient of rats in the western medicine group,the combined Chinese and western medicine group and the high-dose Chinese medicine group were significantly decreased(P<0.05),and the liver coefficient of rats in the middle dose Chinese medicine group was significantly decreased(P<0.05).Compared with the model group,the serum CHO,TG and LDL contents of rats in the western medicine group,the combined Chinese and western medicine group,the high-dose Chinese medicine group and the medium dose Chinese medicine group were significantly decreased(P<0.05),and the H/L was significantly increased(P<0.05).The contents of HDL in serum of rats in the combined Chinese and western medicine group and the high-dose Chinese medicine group were significantly lower than those in the model group(P<0.05).Compared with the model group,the contents of Apo-A1 and Apo-B100 in the serum of rats in the combined Chinese and western medicine group and the high-dose Chinese medicine group were significantly decreased(P<0.05),and the Apo-B100 in the western medicine group and the low-dose Chinese medicine group were significantly decreased(P<0.05).Compared with the model group,the content of MDA in serum of rats in the combined Chinese and western medicine group,the middle dose Chinese medicine group and the high dose Chinese medicine group was significantly decreased(P<0.05),while the activities of SOD and T-AOC were significantly increased(P<0.05).Compared with the model group,the contents of HMG-COA and ACAT in the liver tissue of rats in the combined Chinese and western medicine group and the high-dose Chinese medicine group were significantly decreased(P<0.05).Conclusion:Zhentongji Capsule can effectively reduce the blood lipid level of hyperlipidemic rats,which was closely related to its antioxidation and inhibition of cholesterol synthesis.
