摘要
研制一种高通量自动化的DNA组装仪器,以解决DNA组装过程中通量低、成本高、自动化程度低等问题。通过制作具有高密集度小孔的96孔DNA组装循环加热模块,八连排移液器组件及运动模块,搭建自动化DNA组装仪器;通过软件编写实现温度及移液量的准确控制。经验证DNA组装仪温控性能和移液性能稳定,移液量的重复性为1.23%~7.55%。研制的DNA组装仪可以满足DNA组装实验的移液要求及温度控制要求。
A high-throughput automatic DNA assembly instrument is developed to solve the problems of low throughput,high cost,and low degree of automation in the DNA assembly process.We made a 96-well DNA assembly cycle heating system with high density of holes,eight row pipette assembly and motion module to build automatic DNA assembly instrument.The accurate temperature control and liquid transfer volume was realized through software programming.The stable temperature control performance and pipetting performance of the DNA assembler were verified.The developed DNA assembly instrument can meet the requirements of liquid transfer and temperature control in DNA assembly experiment.
