新型降糖药卡格列净的临床研究进展

Progress in the clinical research of new hypoglycemic drug canagliflozin

当前,全球糖尿病患者人数达到新高,形势严峻。传统降糖药长期应用疗效会降低,且易产生各种不良反应。因此,研发和应用新型降糖药迫在眉睫。新型降糖药卡格列净可通过抑制近端肾小管钠-葡萄糖协同转运蛋白2(sodium-glucose cotransporters 2,SGLT2)介导的葡萄糖重吸收,促进尿糖排泄而发挥降糖作用,临床上用于治疗2型糖尿病(type 2 diabetes mellitus,T2DM)及其合并的心血管疾病、慢性肾脏疾病,疗效显著,安全性高。本文通过对卡格列净的临床疗效和安全性进行综述,为其临床合理应用、T2DM及其并发症治疗提供参考。

At present,the number of patients with diabetes in the world have reached a new high,and the situation is grim.Long-term application effect of traditional hypoglycemic drugs will be reduced,and it is easy to produce various adverse reactions.Therefore,research,development and application of new hypoglycemic drugs are extremely urgent.As a newly marketed hypoglycemic agent,canagliflozin can promote urinary glucose excretion by inhibiting glucose reabsorption mediated by sodium-glucose cotransporters 2(SGLT2)in proximal renal tubules.Clinically,it can be used to treat type 2 diabetes mellitus(T2 DM)and the associated cardiovascular diseases and chronic kidney diseases,which displays a remarkable curative effect with high safety.In this paper,clinical efficacy and safety of canagliflozin were reviewed,so as to provide reference for rational application of canagliflozin to the treatment of T2 DM and related complications.
