高压氧对精索静脉曲张大鼠睾丸组织缺氧诱导因子1α、脂质过氧化物、血管紧张素转化酶的影响及其作用机制

摘要 目的:分析高压氧对精索静脉曲张大鼠睾丸组织缺氧诱导因子1α(HIF-1α)、脂质过氧化物(LPO)、血管紧张素转化酶(ACE)表达的影响及其机制。方法:将雄性SD大鼠随机分为假手术组、模型组和高压氧组;模型组和高压氧组行精索静脉曲张手术造模,假手术组仅手术而不造模;假手术组和模型组大鼠手术或造模成功后正常饲养,高压氧组采用高压氧进行干预治疗;取睾丸计重,行TUNEL法检测睾丸生精细胞凋亡;用免疫印迹和实时荧光定量PCR法检测睾丸组织中HIF-1α、LPO、ACE及其mRNA表达;用ELISA检测试剂盒测定睾丸组织LPO、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、ACE表达水平。结果:高压氧组大鼠左侧、右侧和两侧睾丸质量均明显高于模型组;高压氧组大鼠睾丸细胞凋亡率均明显低于模型组,睾丸组织LPO表达水平明显低于模型组,而SOD、CAT、ACE表达水平均明显高于模型组;高压氧组大鼠睾丸组织HIF-1α、LPO蛋白和mRNA表达均明显低于模型组,ACE蛋白和mRNA水平明显高于模型组。结论:高压氧对雄性SD大鼠精索静脉曲张干预后可有效调节大鼠睾丸组织中HIF-1α、LPO和ACE的表达,进而减少睾丸细胞凋亡。 Objective:To analyze the effect of hyperbaric oxygen on the expression of hypoxia inducible factor-1α(HIF-1α),lipid peroxidation(LPO) and angiotensin converting enzyme(ACE) in male SD rats with varicocele and its mechanism.Methods:SD male rats were divided into a sham operation group,a model group,and a hyperbaric oxygen group by random number table randomly.The model group and the hyperbaric oxygen group received varicocele surgery,while the sham group received surgery without varicocele.Rats in the sham operation group and model group were fed normally after successful operation or modeling,and the hyperbaric oxygen group was treated with hyperbaric oxygen for intervention.The testes were weighed,and the TUNEL method was used to detect the apoptosis of spermatogenic cells.Western blotting method and quantitative real-time PCR were used to detect the expression of HIF-1α,LPO,ACE and their mRNA in the tissues.LPO,superoxide dismutase(SOD),catalase(CAT),and ACE ELISA kits were used to determine the protein expression level of testis tissue.Results:The mean weight of left,right,and bilateral testes in hyperbaric oxygen group were significantly higher than the model group.The testicular cell apoptosis rate of the rats in the hyperbaric oxygen group was significantly lower than the model.The LPO level of rats in the hyperbaric oxygen group was significantly lower than that of the model group,and the levels of SOD,CAT and ACE were significantly higher than those of the model group.The HIF-1α,LPO in the testis of rats in the hyperbaric oxygen group were significantly lower than those in the model group,and the levels of ACE protein and mRNA were significantly higher than those in the model group.Conclusion:The intervention of hyperbaric oxygen on male SD rats with varicocele can effectively regulate the expression of HIF-1α,LPO and ACE in the rat testis and derease testicular cell apoptosis.
