摘要
通过机械破碎和差速离心制备聚丙烯微/纳塑料(PP MPs/NPs)颗粒,以考察其细胞毒作用,采用荧光标记和活细胞成像技术在单核巨噬细胞(RAW 264.7)和肝细胞(HL 7702)两种典型细胞中探索其潜在的毒理学机制。实验结果表明,细胞的摄取/积累水平、毒性和氧化应激(ROS)与颗粒大小和细胞类型具有相关性,潜在的毒理学机制是PP MPs/NPs诱导的ROS导致线粒体损伤和细胞凋亡。本研究提供了涉及PP MPs/NPs对细胞毒理学影响的基本信息。
Polypropylene micro/nanoplastics(PP MPs/NPs) are prepared by mechanical homogenization and differential centrifugation, and their cytotoxic effect and potential toxicological mechanism are investigated by using fluorescence labeling and living cell imaging techniques in two typical cells-mononuclear macrophages(RAW 264.7) and hepatocytes(HL 7702). The experimental results show that cell uptake/accumulation levels, toxicity and oxidative stress(ROS) all have a high correlation with particle size and cell type. The potential toxicological mechanism is that PP MPs/NPs induced ROS leads to mitochondrial damage and apoptosis. This study provides basic information involving the toxicological effects of PP MPs/NPs on cell.
