摘要
为了探究虹鳟传染性胰脏坏死症(Infectious Pancreatic Necrosis,IPN)抗性的遗传基础,本文采用三种二歧性状的分析方法,对来自58个全同胞家族的749尾虹鳟(271尾抗性和478尾易感),注射传染性胰脏坏死病毒后的存活情况进行全基因组关联分析(Genome-Wide Association Study,GWAS)。关联分析结果表明,共鉴定到两个QTN与虹鳟IPN抗性显著关联,位于四号染色体和对应的Scaffold上。以这些检测到的QTNs作为探针,对比虹鳟全基因组信息,筛选得到四个候选基因,它们可能是影响虹鳟IPN抗性的重要功能基因。通过对比三种方法的检测结果,得知GRAMMAR-Lambda法对检测二歧性状QTNs的统计能力略高于SAIGE和GMMAT法,具有更好的统计性能。这些研究结果为虹鳟抗传染性胰脏坏死病毒的遗传选育提供了分子标记,为虹鳟抗传染性胰脏坏死病毒的研究提供了理论基础。
In order to explore the genetic basis of resistance to infectious pancreatic necrosis(IPN)in rainbow trout,a genome wide association study(GWAS)was conducted on 749 rainbow trout(271 resistant and 478 susceptible)from 58 full sib families after injection of infectious pancreatic necrosis virus(IPN)by using three methods for analyzing binary traits.The results showed that two SNP markers were significantly correlated with the anti-infectious pancreatic necrosis virus of O.mykiss,located on chromosomes 14 and the corresponding Scaffolds,respectively.Using these detected QTNs as probes,we compared the whole genome information of O.mykiss,and four candidate genes nearby were found,which may be functional genes affecting the resistance to IPNV in O.mykiss.By comparing the three methods,GRAMMAR-Lambda has a slightly higher statistical ability than SAIGE and GMMAT in detecting QTNs of binary traits and has better statistical performance.These results provide molecular markers for genetic selection of anti-infectious pancreatic necrosis virus in O.mykiss and the molecular basis for further research of the mechanism of infectious pancreatic necrosis virus in O.mykiss.
