归芪藤草膏对外阴慢性单纯性苔藓大鼠肝肾功能及Th1/Th2免疫机制的影响

Effects of Guiqi Tengcao ointment on liver and kidney function and Th1/Th2 immune mechanism in vulvar chronic lichen simplex rats

目的探讨归芪藤草膏对外阴慢性单纯性苔藓大鼠外阴组织学、肝肾功能及Th1/Th2免疫机制的影响。方法取40只雌性SD大鼠,随机取10只作为空白组,其余大鼠采用0.5%二甲基苯并蒽丙酮液联合机械刺激方法建立外阴慢性单纯性苔藓病变。将造模成功大鼠随机分为归芪藤草膏组和模型组各13只,归芪藤草膏组给予归芪藤草膏外涂外阴治疗4周,模型组给予蒸馏水外涂外阴治疗4周,空白组不给予任何干预。观察各组大鼠外阴皮肤变化,ELISA法检测外周血谷丙转氨酶(AST)、谷草转氨酶(ALT)、尿素氮(BUN)、血肌酐(SCr)、白细胞介素-2(IL-2)、干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)及白细胞介素-6(IL-6)水平。结果模型组大鼠外阴皮肤颜色较红,触诊皮肤粗糙、弹性差;归芪藤草膏组大鼠干预4周后皮肤颜色较模型组明显好转,触诊皮肤光滑有弹性,皮肤外观与空白组大致相同。模型组大鼠血清AST、BUN、IL-2、IL-4、IL-6水平均明显高于空白组(P均<0.05),血清IFN-γ水平明显低于空白组(P<0.05);归芪藤草膏组血清ALT、AST、BUN、IL-2、IL-4、IL-6水平均明显低于模型组(P均<0.05),血清IFN-γ水平明显高于模型组(P<0.05);归芪藤草膏组血清ALT、AST、IL-2和IFN-γ水平均明显低于空白组(P均<0.05),血清BUN、IL-4和IL-6水平均明显高于空白组(P均<0.05);各组间血清SCr水平比较差异均无统计学意义(P均>0.05)。结论归芪藤草膏能显著减轻外阴慢性单纯性苔藓大鼠的皮肤损伤,机制可能与调节肝肾功能及Th1型和Th2型细胞因子水平有关。

Objective It is to investigate the effects of Guiqi Tengcao ointment on vulvar histology,liver and kidney function and Th1/Th2 immune mechanism in vulvar chronic lichen simplex rats.Methods Forty female SD rats were selected,in which 10 rats were randomly selected as blank group,and the remaining rats were treated with 0.5%dimethylbenzanthracene acetone solution combined with mechanical stimulation to establish the lesions of vulvar chronic lichen simplex.The successful modeled rats were randomly divided into Guiqi Tengcao ointment group and model group,13 rats in each group.The Guiqi Tengcao ointment group was treated with Guiqi Tengcao ointment by vulva external application for 4 weeks,the model group was treated with distilled water by vulva external application for 4 weeks,and the control group was not intervened for 4 weeks.The changes in the vulva skin of the rats in each group were observed,and the levels of peripheral blood alanine aminotransferase(AST),aspartate aminotransferase(ALT),blood urea nitrogen(BUN),serum creatinine(SCr),interleukin-2(IL-2),interferon-γ(IFN-γ),interleukin-4(IL-4)and interleukin-6(IL-6)were detected by ELISA.Results The vulva skin color of the rats in the model group was redder,with rough skin and poor elasticity in palpation;after 4 weeks of intervention,the skin color of the rats in the Guiqi Tengcao ointment group was significantly improved compared with the model group,the skin was smooth and elastic in palpation,and the skin appearance was roughly the same as that in the blank group.The levels of serum AST,BUN,IL-2,IL-4 and IL-6 in the model group were significantly higher than those in the blank group(P<0.05),and the serum IFN-γlevel was significantly lower than that in the blank group(P<0.05);the levels of serum ALT,AST,BUN,IL-2,IL-4 and IL-6 in the Guiqi Tengcao ointment group were significantly lower than those in the model group(all P<0.05),and the level of serum IFN-γwas significantly higher than that in the model group(P<0.05);the levels of serum ALT,AST,IL-2 and IFN-γin the Guiqi Tengcao ointment group were significantly lower than those in the blank group(all P<0.05),and the levels of serum BUN,IL-4 and IL-6 were significantly higher than those in the blank group(all P<0.05),there was no significant difference in serum SCr levels among the groups(all P>0.05).Conclusion Guiqi Tengcao ointment can significantly improve the vulvar skin condition of rats with vulvar lichen simplex chronicus.Its mechanism may be related to regulating the level of liver and kidney function and related inflammatory factors in rats.
