结直肠癌组织中T-框蛋白5表达与临床病理特征及预后的关系

Expression of T-box protein 5 in colorectal cancer tissues and its association with clinicopathological characteristics and prognosis of colorectal cancer

目的 探讨结直肠癌组织中T-框蛋白5表达与临床病理特征及预后的关系。方法 选取2020年2月至2021年2月间锦州市中心医院收治的180例结直肠癌患者,分析结直肠癌组织中TBX5表达与临床病理特征的关系,统计分析人结肠癌细胞株中、各组HT29细胞中TBX5 mRNA和蛋白表达水平。结果 180例患者中,TBX5表达阳性44例,阳性率为24.4%,其中浸润深度T1、T2、T3、T4患者的TBX5表达阳性率逐渐降低,差异均有统计学意义(均P<0.05)。有淋巴结转移、神经侵犯患者的TBX5表达阳性率分别低于无淋巴结转移、无神经侵犯患者,差异均有统计学意义(均P<0.05)。SW620细胞、LOVO细胞、HCT116细胞、SW480细胞、HT29细胞中TBX5 mRNA和蛋白表达水平均逐渐降低,差异均有统计学意义(均P<0.05)。TBX5转染组HT29细胞中TBX5 mRNA和蛋白表达水平均高于阴性对照组、空白对照组,差异均有统计学意义(均P<0.05)。结论 结直肠癌组织中T-框蛋白5表达与浸润深度、淋巴结转移、神经侵犯及预后的关系密切。

Objective To investigate the relationship between the expression of T-box protein 5(TBX5) in colorectal cancer tissues and clinicopathological characteristics and prognosis of colorectal cancer. Methods A total of 180 patients with colorectal cancer were selected at Jinzhou Central Hospital from February 2020 to February 2021. The relationship between expression of TBX5 and clinicopathological characteristics of colorectal cancer tissues was analyzed. The expression of TBX5 mRNA and protein was statistically analyzed in human colon cancer cell lines and HT29 cells for each group. Results Among the 180 patients, 44 were positive for TBX5 expression, with a positive rate of 24.4%. Among them, the positive rate of TBX5 expression gradually decreased in patients with depth of invasion T1, T2, T3, and T4(P<0.05). The positive rates of TBX5 expression were lower in patients with lymph node metastasis and nerve invasion than those without lymph node metastasis and nerve invasion(all P<0.05). TBX5 mRNA and protein expression levels gradually decreased in SW620 cells, LOVO cells, HCT116 cells, SW480 cells, and HT29 cells(all P<0.05). The expression levels of TBX5 mRNA and protein were higher in HT29 cells in the TBX5 transfection group than those in the negative control group and the blank control group(all P<0.05). Conclusion The expression of T-box protein 5 in colorectal cancer tissues is closely associated with the depth of invasion, lymph node metastasis, nerve invasion and prognosis.
