^(131)I标记复合靶向基因载体对人脑胶质瘤U251细胞恶性生物学行为的影响

Effect of ^(131)I labeled compound targeting gene vector on malignant biological behavior of human glioma U251 cells

摘要 目的考察^(131)I标记复合靶向基因载体对人脑胶质瘤U251细胞恶性生物学行为的影响。方法选取U251细胞为研究对象,随机分为对照组与观察组两个组别。对照组细胞转入空白载体,观察组细胞中则载入^(131)I标记复合靶向基因载体。对两组细胞的载体转染情况、细胞迁移、侵袭能力、黏附能力及相关基因表达情况进行分析。结果^(131)I的纳米载体最终标记率为71%~85%,放化纯度为98.6%~99.2%;观察组U251细胞的增殖速率明显低于对照组,差异具有统计学意义(P<0.05);观察组中U251细胞转移、侵袭能力明显低于对照组,差异具有统计学意义(P<0.05);观察组中U251细胞的黏附能力明显低于对照组,差异具有统计学意义(P<0.05);观察组细胞中与肿瘤转移相关的基因明显下调。结论^(131)I标记复合靶向基因载体能够在一定程度上改善人脑胶质瘤U251细胞恶性生物学行为,抑制U251细胞的迁移及侵袭,降低其黏附力,对相关疾病的治疗具有一定的指导意义。 Objective To investigate the effect of^(131)I labeled compound targeting gene vector on the malignant biological behavior of human glioma U251 cells.Methods U251 cells were randomly divided into control group and observation group.The cells in the control group were transferred into blank vector,and the cells in the observation group were loaded with^(131)I labeled compound targeted gene vector.The vector transfection,cell migration,invasion,adhesion and related gene expression of the two groups were analyzed.Results The final labeling rate of^(131)I nano carrier is 71%~85%,and the radiochemical purity is 98.6%~99.2%;The ability of cell metastasis and invasion in the observation group was sig⁃nificantly lower than that in the control group(P<0.05);The adhesion ability of cells in the observation group was sig⁃nificantly lower than that in the control group(P<0.05);The genes related to tumor metastasis were significantly down regulated in the observation group.Conclusion^(131)I labeled compound targeting gene vector can improve the malignant biological behavior of human glioma U251 cells to a certain extent,inhibit the migration and invasion of U251 cells,and reduce their adhesion.It had certain guiding significance for the treatment of related diseases.
