摘要
试验旨在研究使用杏鲍菇菌糠制备低聚木糖饲料的工艺。试验以杏鲍菇菌糠为原料,低聚木糖产量为指标,采用单因素试验法研究杏鲍菇菌糠预处理条件及酶解条件对低聚木糖产量的影响,并对低聚木糖产量影响较大的因素通过正交试验法进行优化。结果显示:适宜的预处理工艺为:预处理温度50℃、H_(2)O_(2)3%、Na_(2)CO_(3)4%、液料比2.5 mL/g、预处理时间3 h。适宜的酶解工艺为:木聚糖酶400 U/g、纤维素酶400 U/g、甘露聚糖酶400 U/g、液料比4 mL/g、酶解温度50℃、酶解初始pH值6.0、酶解时间4 h。此条件下,酶解物料中低聚木糖产量达97.67 mg/g。研究表明,该工艺为杏鲍菇菌糠的高效利用提供了新思路。
The experiment was to study the technology of preparing xylooligosaccharide feed by using Pleurotus eryngii mushroom bran. Using Pleurotus eryngii mushroom bran as raw material and xylooligosaccharide yield as index, single factor experiment was used to study the effect of pretreatment conditions and enzymatic hydrolysis conditions on xylooligosaccharide yield, and the factors with great influence on xylooligosaccharide yield were optimized by orthogonal experiment. The results showed that the appropriate pretreatment process was pretreatment temperature 50 ℃, H_(2)O_(2)3%,Na_(2)CO_(3)4%, liquid-solid ratio 2.5 mL/g, pretreatment time 3 h. The suitable enzymatic hydrolysis process was xylanase400 U/g, cellulase 400 U/g, mannanase 400 U/g, liquid-solid ratio 4 mL/g, enzymatic hydrolysis temperature 50 ℃, initial pH value 6.0, enzymatic hydrolysis time 4 h. Under these conditions, the yield of xylooligosaccharide in the enzymatic hydrolysis material reached 97.67 mg/g. The study indicates that the technology provides a new idea for efficient utilization of Pleurotus eryngii mushroom bran.
