细胞松弛素D对小鼠乳腺腺癌细胞系(MA782/5S-8102)细胞的作用

摘要 本文研究了细胞松弛素D(CD)对小鼠乳腺腺癌细胞系(MA782/5S-8102)细胞的作用,统计了在0.4微克/毫升CD作用下的多核细胞形成率,并与HEp2细胞及正常婴兔肾原代培养细胞进行比较。结果表明:CD处理48小时后,肿瘤细胞形成大量的多核,≥3核的细胞数在MA782/5S-8102细胞中为37.05%,在HEp2细胞中为18.37%。正常婴兔肾原代培养细胞经同样处理后≥3核的细胞数只有0.059%。肿瘤细胞多核形成的比例会随CD处理时间的延长而显著增高,并出现排核现象。CD处理96、144小时后的多核细胞数,MA782/5S-8102细胞分别为47.90%,53.12%,HEp2细胞分别为43.94%,51.94%,而正常婴兔肾原代培养细胞则仅为0.26%和0.39%,亦未见排核现象,肿瘤细胞和正常细胞对CD的不同反应,有可能作为鉴别肿瘤细胞与正常细胞的一个简便易行的工具。 MA782/5S-8102 cell line derived from mouse mammary adenocarcinoma was examined for its response to cytochalasin D(CD).The multinucleation of cells in the presence of CD at concentration of 0.4μg/ml was measured.Neoplastic HEp2 cell line and normal primary culture derived from the baby rabbit kidney were also examined as positive and negative controls.The results demonstrated that MA782/5S-8102 and HEp2 cells extensively multinuclealed in response to CD,whereas normal primary baby rabbit kidney culture cells did not multinucleate.After 48h exponsed to CD,the mean percentage of multinucleated cells(≥3 nuclei/cell)was:MA782/5S-8102 cells,37.05%;HEp2 cells,18.37%.And as the duration of exponse time to CD increased,the multinucleated cells increased in number also.Untill 144h,the number of multinucleated cells in MA782/5S-8102 and HEp2 was 53.12%and 51.94%respectively,whereas only 0.39%of cells in normal primary baby rabbit kidney cultures exhibited multinucleation.From above observations,it clearly shows that the uncontrolled nuclear division is a characteristic of neoplastic cells different from normal cells.These rcsults suggest that the different response of tumor cells and normal cells to CD may be a useful tool for distinguishing normal cells from neoplastic cells in vitro.
