烙铁头蛇毒血小板聚素(TMVA)对动物体内血小板的聚集与止血功能

STUDY OF PLATELETS AGGREGATION AND THE EFFECT OF HEMOSTASIE IN VIVO BY TMVA

兔子实验表明静脉注射TMVA后血小板数量降低70%以上,3小时后回升,12小时左右接近正常。经注射TMVA后PF_(3),PF_(4)活性增强,出血时间缩短,全血凝固时间也略有缩短,在不同时间内取心脏血测定结果证明血小板聚集功能降低,低剂量组呈波浪状变化,24小时尚未恢复正常。经对给药后兔子24小时及30天的病理切片检查发现仅个别动物有血栓,血小板聚集和红细胞沉积。小白鼠腹腔注射TMVA后,出血时间也明显缩短,而大剂量组5小时后出血时间略有延长。

Results of experiment on rabbit indicate:after the injection of TMVA intravenously,the counting of blood platelets obviously dropped to 30%of the original.It gradually raised after three hours and it became normal about 12 hours afterwards.No thrombus discovered from animals killed from 24 hours to 30 days.Aggregation of platelets and deposition of red blood cell appeared in only a few of those animals.Blood taken from hearts at various intervals for testing experiments of platelets aggregation results showed that obvious decrease in aggregation function of platelets through exhaustion.It was impossible for all of them return to normal 24 hours afterwards except those groups treated with lower dosage.Whole blood coagulation time:PF3;PF4 activity raised,Shightly shorter at 3 hours bleeding time shorter within 30 minutes no other obvious changes appeared.Determination of platelets aggregation function at various stages showed obvious decrease.Through exhaustion of the aggregation function,except that wave-like variations appeared for those treated with low dosage.Mouse injected with TMVA.The bleeding time obviously became shorter within 30 minutes,those groups treated with high dosage prolonged after 5 hours.
