电针“太冲”对自发性高血压大鼠海马MDA、SOD、GSH-Px含量的影响 被引量:3

Effect of Electroacupuncture on"Taichong"on Contents of MDA,GSH-Px and SOD in Hippocampus of Spontaneously Hypertensive Rats

摘要 目的:通过观察电针“太冲”对自发性高血压大鼠(spontaneously hypertensive rats,SHR)海马损伤的改善情况,探讨电针治疗高血压的中枢机制。方法:按照随机数字表法将12只雄性SHR大鼠分为电针组与模型组2组各6只,6只雄性WKY大鼠为空白组,适应性喂养1周后电针组用电针仪分别连接双侧“太冲”穴,电流强度1 mA,频率2 Hz,留针20 min连续针刺14 d,每日1次。实验第15天,HE染色法观察海马组织的形态结构,并测定海马组织中丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)含量。结果:与空白组比较,模型组HE染色明显变浅,出现细胞溶解碎裂、核固缩、空泡样变性等;与模型组比较,电针组细胞染色加深,核仁较清晰,细胞结构基本正常;与空白组比较,模型组的MDA含量显著提升(P<0.01),SOD、GSH-Px含量显著降低(P<0.01);与模型组比较,电针组MDA含量明显下降(P<0.05),SOD、GSH-Px含量明显上升(P<0.05)。结论:电针“太冲”穴能通过调节SHR大鼠海马中MDA、SOD和GSH-Px的表达水平,抑制氧化反应,减轻氧自由基对SHR大鼠海马组织的损伤,这可能是电针“太冲”穴对高血压的中枢保护机制之一。 Objective:To explore the central mechanism of electroacupuncture(EA)on"Taichong"(LR3)in the treatment of hypertension by observing the improvement of hippocampal injury in spontaneously hypertensive rats(SHRs).Methods:12 male SHR rats were divided into 2 groups by random number table method:the EA group and the model group,with 6 rats in each group.Six male WKY rats served as the blank group.After one week of acclimatization,the EA group was intervened with EA on bilateral“Taichong”(LR3)once a day for 14 days(1mA,2Hz,20min).On the 15th day,we collected hippocampus samples.The morphology of hippocampus was observed by HE staining,and the contents of MDA,SOD and GSH-Px in hippocampus were detected.Results:Compared with the blank group,the staining was significantly lighter in the model group,with cell lysis and fragmentation,nuclear pyknosis and vacuolar degeneration.Compared with the model group,the cell staining of the EA group was deepened,the nucleolus was clearer,and the cell structure was basically normal.Compared with the blank group,MDA content in model group increased significantly(P<0.01),while SOD and GSH-Px contents decreased significantly(P<0.01).Compared with the model group,MDA content in EA group decreased significantly(P<0.05),while SOD and GSH-Px content increased significantly(P<0.05).Conclusion:EA on LR3 can regulate the expression of MDA,SOD and GSH-Px in the hippocampus of SHR rats,inhibit the oxidative reaction,and reduce the damage of oxygen radicals to the hippocampus of SHR rats,which may be one of the central protective mechanisms of EA on LR3 in treating hypertension.
