NNMT在大肠腺癌与大肠腺瘤的表达对比及对大肠腺癌短期生存的影响

Expression of NNMT in Colorectal Adenocarcinoma and Colorectal Adenoma and Its Influence on Short-term Survival of Colorectal Adenocarcinoma

目的:探究与分析烟酰胺N-甲基转移酶(NNMT)在大肠腺癌与大肠腺瘤的表达差异及对大肠腺癌短期生存的影响。方法:选取2017年2月-2019年2月福建省立医院南院外科手术或内镜下取材的正常大肠黏膜组、大肠腺瘤组及大肠腺癌组,各50例。对比NNMT mRNA及其蛋白在正常大肠黏膜组、大肠腺瘤组及大肠腺癌组组织中相对表达量,分析NNMT表达与大肠腺癌临床病理因素的关系,对比不同NNMT表达情况下大肠腺癌患者短期生存期。结果:大肠腺瘤组、大肠腺癌组NNMT mRNA及其蛋白表达量均高于正常大肠黏膜组,大肠腺癌组NNMT mRNA及其蛋白表达量均高于大肠腺瘤组,差异均有统计学意义(P<0.05)。不同NNMT表达情况大肠癌患者的组织学分级比较,差异有统计学意义(P<0.05)。大肠腺癌患者均接受为期3年的随访,NNMT阳性表达患者3年生存率为50.00%(14/28),低于NNMT阴性表达患者的90.91%(20/22),差异有统计学意义(χ^(2)=9.475,P=0.002)。结论:NNMT在大肠腺瘤与大肠腺癌组织中的表达存在明显的差异性,NNMT阳性表达患者的短期生存率较低。

Objective:To explore and analyze the difference of nicotinamide N-methyltransferase (NNMT) expression in colorectal adenocarcinoma and colorectal adenoma and its influence on the short-term survival of colorectal adenocarcinoma.Method:From February 2017 to February 2019,the normal colorectal mucosa group,the colorectal adenoma group and the colorectal adenocarcinoma group obtained by surgery or endoscopy group of the South Hospital of Fujian Provincial Hospital were selected,50 cases in each group.The relative expression levels of NNMT mRNA and its protein in normal colorectal mucosa group,colorectal adenoma group and colorectal adenocarcinoma group tissue were compared,the relationship between NNMT expression and colorectal adenocarcinoma clinicopathological factors was analyzed,and the short-term lifetimes of colorectal adenocarcinoma patients with different NNMT expression were compared.Result:The expression levels of NNMT mRNA and protein in the colorectal adenoma group and colorectal adenocarcinoma group were higher than those in the normal colorectal mucosa group,and the NNMT mRNA and protein expressions in the colorectal adenocarcinoma group were higher than those in the colorectal adenoma group,the differences were statistically significant (P<0.05).Comparison of histological grades of colorectal adenocarcinoma patients with different NNMT expression,the difference was statistically significant (P<0.05).Colorectal adenocarcinoma patients were followed up for 3 years,the 3-year survival rate of patients with positive NNMT expression was 50.00% (14/28),which was lower than 90.91% (20/22) of patients with negative NNMT expression,the difference was statistically significant (χ^(2)=9.475,P=0.002).Conclusion:There are significant differences in the expression of NNMT in colorectal adenoma and colorectal adenocarcinoma,and the short-term survival rate of patients with positive NNMT expression is low.
