摘要
本文以明胶为载体材料,绿原酸作为模型药物,采用乳化-化学交联法制备绿原酸微球。以产率、包封率、平均粒径为评价指标,通过响应曲面法优化制备工艺。得到的最佳制备工艺为:明胶浓度为0.23g·mL^(-1)、水油体积比为1∶5.6(mL∶mL)、搅拌速度为1220r·min^(-1)。该条件下制备的微球表面光滑圆整,大小均匀,分散性好,平均粒径为36μm,产率为99.5%。通过紫外分光光度法测得包封率为89.5%。说明该方法制备的绿原酸微球稳定性好、工艺简单、产率高,具有良好的应用前景和推广价值。
Using gelatin as carrier material and chlorogenic acid as model drug,chlorogenic acid microspheres were prepared by emulsification-chemical cross-linking method.Taking yield,encapsulation efficiency and average particle size as evaluation indicators,the preparation process was optimized by response surface method.The best preparation process was obtained as follows:the gelatin concentration was 0.23g·mL^(-1),the water-oil volume ratio was 1∶5.6(mL∶mL),and the stirring speed was 1220r·min^(-1).The microspheres prepared under these conditions were smooth and round in surface,uniform in size and good in dispersibility,with an average particle size of 36μm and a yield of 99.5%.The encapsulation efficiency was determined to be 89.5%by UV spectrophotometry.It showed that the chlorogenic acid microspheres prepared by this method have good stability,simple process and high yield,and have good application prospects and promotion value.
