摘要 为研究AsWOX9基因参与调控大蒜鳞芽休眠的功能和分子机理,从大蒜品种“二水早”叶片中成功克隆出AsWOX9基因,并对其进行生物信息学及在大蒜各器官中的表达分析。结果表明,AsWOX9的cDNA序列全长为570 bp,编码189个氨基酸;AsWOX9蛋白的分子式为C_(939)H_(1488)N_(276)O_(296)S_(12),相对分子质量为21764.54,等电点8.47,为亲水性蛋白,且是没有信号肽、剪切位点和跨膜结构的非分泌蛋白。AsWOX9蛋白存在homeobox结构域,属于homeodomain型家族基因,定位于细胞核中。AsWOX9与芦笋的WUSCHEL-relat⁃ed homeobox 8亲缘关系最近。进一步的qRT-PCR结果显示,AsWOX9基因在大蒜组织器官中广泛表达,尤以叶中表达量最高。本研究结果可为AsWOX9基因的功能研究奠定理论基础。 To study the function and molecular mechanism of AsWOX9 gene involved in regulating the dormancy of garlic scales,AsWOX9 gene was successfully cloned from the leaves of garlic variety“Er⁃shuizao”,and its bioinformatics and expression in various organs of garlic were analyzed.The results showed that the total length of cDNA sequence of AsWOX9 was 570 bp,encoding 189 amino acids.The chemical for⁃mula of AsWOX9 protein was C_(939)H_(1488)N_(276)O_(296)S_(12),the relative molecular mass was 21764.54,and the isoe⁃lectric point was 8.47,which was a hydrophilic protein.This protein was a non⁃secreted protein without signal peptide,cleavage site and transmembrane structure.AsWOX9 protein had a homeobox domain,belonged to the homeodomain type family gene,and was located in the nucleus.It had the closest genetic relationship with the WUSCHEL-related homeobox 8 of asparagus.qRT-PCR results showed that AsWOX9 gene was widely ex⁃pressed in tissues and organs of garlic,with the highest expression in leaves.This results could provide theo⁃retical bases for the study of gene function of AsWOX9.
