摘要
为研究NR3C2基因与鸡繁殖性能的相关性,本试验采用PCR-RFLP方法检测了鸡盐皮质激素受体(NR3C2)基因在汶上芦花鸡、济宁百日鸡和莱芜黑鸡3个地方鸡种和1个引进高产蛋鸡品种海兰褐中的多态性,结果在4个群体中检测到NR3C2基因3714 nt处发生点突变G→A,基因型GG在3个地方鸡种中表现为劣势,而在高产蛋鸡海兰褐中表现为优势基因型,频率为88.84%,优势等位基因G频率为93.93%。该位点各基因型与392只莱芜黑鸡4个繁殖性状的关联分析结果表明3714位点与43W产蛋量(P=0.0467)、44W产蛋量(P=0.0486)和45W产蛋量(P=0.0499)均表现出显著相关,且GG基因型个体的开产日龄最早,比GA基因型个体平均早1.17 d。因此,GG基因型可以作为地方鸡种繁殖性状的分子标记,在地方鸡种遗传改良过程中加以利用。
In order to study the correlation between NR3C2 gene and chicken reproductive performance,the PCR⁃RFLP method was used to detect the polymorphism of NR3C2 gene in three local chicken breeds of Wenshang barred chicken,Jining Bairi chicken and Laiwu black chicken and the introduced Hy⁃Line brown chicken with high yield.The results showed that point mutation G→A was detected at 3714 nt of NR3C2 gene in the four populations.The frequency of genotype GG was lower in three local chicken breeds,but it was pre⁃dominant in Hy⁃Line brown chicken with the frequency as 88.84%and the G allele frequency as 93.93%.The results of correlation between each genotype of this locus and four production traits of 392 Laiwu black chicken showed that the locus at 3714 nt had significant association with egg production of 43W(P=0.0467)、44W(P=0.0486)and 45W(P=0.0499);the GG genotype individuals had the earliest birth age,with were 1.17 days earlier than that of GA genotype individuals on average.Therefore,the GG genotype could be used as a molecular marker for reproductive traits,and used in the process of genetic improvement of local chicken breeds.
