摘要
目的评价一种两栖类动物皮肤分泌物中的多肽RanacyclinE(氨基酸序列SAPRGCWTKSYPPKPCK)的丝氨酸蛋白酶抑制活性。方法RanacyclinE由Fmoc固相化学合成后经基质辅助激光解析质谱鉴定,测定胰蛋白酶抑制常数Ki。结果RanacyclinE的酶抑制常数Ki值为95.96μM。结论天然生物肽RanacyclinE是一种潜在的丝氨酸蛋白酶抑制剂。
OBJECTIVE Inhibitory activity evaluation of a short peptide from amphibian skin secretion,RanacyclinE(amino acid sequence SAPRGCWTKSYPPKPCK),and assay of its inhibitory constant Ki value.METHODS RanacyclinE was synthsized by Fmoc solid phase peptide synthesis(SPPS)and identified by MALDI-TOF MS.Inhibitory constant Ki value was calculated by Grafit@software.RESULTS Inhibitory constant Ki value was 95.96μM.CONCLUSION Natural biological peptide RanacyclinE is a potential serine protease inhibitor.
