化妆品中人表皮生长因子的UPLC-Q-TOF-MS测定方法和量化确证策略

摘要 建立UPLC-Q-TOF-MS测定化妆品中EGF的方法。冻干粉、水和啫喱化妆品提取、过滤后以0.1%甲酸-0.1%甲酸乙腈为流动相,经C_(18)色谱柱分离,ESI源正离子多反应模式外标法定量。在0.5~10μg/mL内线性关系良好,R^(2)大于0.9973,检出限0.2μg/mL,回收率96.6%~116.6%,RSD小于5.0%(n=6)。该方法简便、准确。提出化妆品中EGF的量化确证策略。联用互不干扰方法,在符合要求下对特征分别赋分,以总分进行不同程度的肯定。该策略可提供化妆品中物质确证的参考方案。 An analysis method for determination of EGF in cosmetics by UPLC-Q-TOF-MS was established.Lyophilized powder,water and gel cosmetics were soluted in 0.2%BSA or 0.4%BSA NaCl solution,and filtered with PTFE millipore membrane.An UPLC method was applied with the C_(18)column by a gradient elution using 0.1%formic acid aqueous solution-0.1%formic acid ACN solution as the mobile phase,EIS in positive ion mode was used in MRM mode,and EGF quantified with external standard method.The results showed a good linear relationship in the corresponding concentration range of 0.5~10μg/mL with a sqaredcorrelation coefficient great than 0.9973,and the limit of detection(LOD)was 0.2μg/mL,and the recovery rate was in range of 96.6%~116.6%with RSD no more than 5.0%(n=6).This method was efficiency and accurate.A strategy for confirming of EGF in cosmetics was also introduced.Combined with orthogonal analysis methods,the detection features meet the acceptable criteria were scored respective,and objective was determined by the total score in deferent degrees.The strategy was scientific and effective,and can provide a solution for the confirmation of substances in cosmetics.
