摘要
在全球新冠肺炎大流行的背景下,迫切需要快速、准确的现场诊断检测来及时识别感染者.现阶段病毒检测的金标准是逆转录实时定量聚合酶链式反应(RT-qPCR),然而这种方法需要特定且昂贵的设备、专业的分析人员以及4~8 h的处理时间.环介导等温扩增技术(LAMP)因其灵敏度高、特异性强、快速、成本低等优点,作为qPCR的一种替代方法,已广泛应用比色法、实时监测法、侧流层析法、微流控芯片、生物传感器等方法来检测新型冠状病毒(SARS-CoV-2).在LAMP检测方法经过样品处理简单化、分析智能化等改进之后,增强型LAMP为现场诊断SARS-CoV-2提供了一种新思路.
In the context of the global COVID-19 pandemic,there is an urgent need for a rapid and accurate on-site diagnostic test to identify infected people in a timely manner.The gold standard for virus detection at this stage is reverse transcription real-time quantitative polymerase chain reaction(RT-qPCR),but this method requires specific and expensive equipment,professional analysts,and a processing time of 4~8 hours.As an alternative method of qPCR,loop-mediated isothermal amplification technology(LAMP)has been widely used in colorimetry,real-time monitoring,lateral flow chromatography,microfluidic chips,biosensors and other methods to detect novel coronavirus(SARS-CoV-2)because of its high sensitivity strong specificity,high speed and low cost.After the improvement of the LAMP detection method,such as sample processing simplification and intelligent analysis,the enhanced LAMP provides a new idea for on-site diagnosis of SARS-CoV-2.
