摘要 试验旨在通过超声波辅助酶法提取杜仲叶中的总黄酮。试验在单因素试验的基础上,利用响应面法优化杜仲叶黄酮提取工艺条件。结果显示:在酶加量10.27 mg、液料比28.96 mL/g、超声时间15.59 min、酶解时间75.14 min条件下,总黄酮得率为3.376%。对最优条件修正后,总黄酮得率为3.295%,理论值与实际值的拟合性较好。研究表明,利用超声波法和酶解法相结合的方法提取杜仲叶中总黄酮可获得较高的总黄酮得率。 The study was to extract total flavonoids from Eucommia ulmoides leaves by ultrasonic assisted enzymatic method.On basis of single factor experiment,response surface method was used to optimize extraction process of flavonoids from Eucommia ulmoides leaves.The results showed that under conditions of enzyme dosage of 10.27 mg,liquid to solid ratio of 28.96 mL/g,ultrasonic time of 15.59 min and enzymatic hydrolysis time of 75.14 min,the yield of total flavonoids was 3.376%,and the yield was 3.295%after the modification of optimal conditions.The fitting between theoretical value and actual value was good.The experiment indicates that the extraction of total flavonoids from Eucommia ulmoides leaves by ultrasonic method and enzymatic hydrolysis method can obtain higher yield.
