摘要
目的:研究莪术醇提物乳膏通过Janus激酶/信号转导和转录激活因子(JAK/STAT3)通路对银屑病大鼠皮肤病理改变的影响。方法:选取48只大鼠(雌雄各半)随机分成4组,即空白对照组、银屑病组、莪术醇提物乳膏(1 g/kg)组、莪术醇提物乳膏(2g/kg)组。建立银屑病大鼠模型,建模成功后,空白对照组、银屑病组不做处理,莪术醇提物乳膏(1 g/kg)组、莪术醇提物乳膏(2 g/kg)组每天给药1次。最后1次给药后2 d观察大鼠变化,采集大鼠背部皮肤组织进行苏木素-伊红(HE)染色,对比分析各组大鼠的皮肤病理改变情况。结果:银屑病组、莪术醇提物乳膏(1 g/kg)组及莪术醇提物乳膏(2 g/kg)组大鼠的表皮厚度、炎症细胞浸润数量、Baker病理评分、白细胞介素-17(IL-17)、IL-22、IL-23、γ-干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)表达水平、IL-6受体/JAK/STAT3(IL-6R/JAK/STAT3)通路因子mRNA表达水平均明显高于空白对照组,P<0.05。银屑病组的上述评价指标均明显高于两个莪术醇提物乳膏组。此外,莪术醇提物乳膏(2 g/kg)组的上述评价指标均明显低于莪术醇提物乳膏(1 g/kg)组,P<0.05。结论:莪术醇提物乳膏对银屑病皮肤病理改变有明显改善作用,其作用机制可能与细胞因子以及免疫表达相关。
Objective:To study the effect of Zedoary Zedoary ethanol extract cream on skin pathological changes in psoriasis rats through Janus kinase/signal transduction and activator of transcription(JAK/STAT3)pathway.Methods:Forty-eight rats(half male and half female)were randomly divided into 4 groups:blank control group,psoriasis group,Zedoary zedoary ethanol extract cream(1 g/kg)group,Zedoary ethanol extract cream(2g/kg)group.The psoriasis rat model was established.After successful modeling,the blank control group and psoriasis group were not treated,and the curcuma zedoary alcohol extract cream(1 g/kg)group and Curcuma zedoary alcohol extract cream(2 g/kg)group were adm inistered once a day.T wo days after the last dose,the changes of rats were observed,and the skin tissues of rats were collected for hematoxylin and eosin(HE)staining,and the skin pathological changes of rats in each group were compared and analyzed.Results:Psoriasis group,Curcuma zedoary ethanol extract cream(1 g/kg)group,and Curcuma zedoary ethanol extract cream(2 The epidermal thickness,inflammatory cell infiltration number,Baker pathological score,interleukin-17(IL-17),IL-22,IL-23,interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α)expression levels,IL-6 receptor/JAK/STAT3(IL-6R/JAK)of rats in g/kg group/STAT3)pathway factors mRNA expression levels were significantly higher than those in blank control group,P<0.05.The above evaluation indexes in psoriasis group were significantly higher than those in two curcuma alcohol extract cream groups.In addition,the above evaluation indexes of Curcuma Zedoary ethanol extract cream(2 g/kg)group were significantly lower than those of Curcuma Zedoary ethanol extract cream(1 g/kg)group,P<0.05.Conclusion:Zedoary Turmeric alcohol extract cream can significantly improve the pathological changes of psoriasis skin,and its mechanism may b e related to the expression of cytokines and immune.
