HPLC法测定邓氏清毒饮中绿原酸的含量

Determination of chlorogenic acid in Deng’s Qingdu drink by HPLC

目的:对邓氏清毒饮用于新型冠状病毒肺炎(新冠肺炎)防控中的可行性进行综合分析,并根据分析结果采用高效液相色谱法(HPLC)对邓氏清毒饮中活性成分绿原酸的含量进行测定。方法:根据邓氏清毒饮的处方解析、药材化学成分、现代药理学机制等综合分析将其用于新冠肺炎防控中的可行性。采用HPLC对邓氏清毒饮中活性成分绿原酸的含量进行测定,测定条件:色谱柱:Agilent ZORBAX SB-C18(4.6 mm×250 mm,5-Micron),流动相:乙腈-0.1%磷酸(10:90),流速:1.0 mL/min,柱温:25℃,检测波长:326 nm。结果:从处方解析、药效活性成分、药理作用机制等方面对邓氏清毒饮进行综合分析,均能在一定程度上对其抗新冠肺炎作用进行解释。经HPLC测定,绿原酸在13.91~278.25μg/mL范围内线性关系良好(r=0.9999),平均回收率为100.03%﹝相对标准偏差(RSD)=1.23%﹞。结论:邓氏清毒饮可用于新冠肺炎的防控,文中所建立的含量测定方法可用于测定邓氏清毒饮中绿原酸的含量。

Objective:To comprehensively analyze the feasibility of Deng’s Qingdu decoction in the prevention and control of COVID-19,and to determine the content of chlorogenic acid in Deng’s Qingdu Decoction by high performance liquid chromatography(HPLC)according to the analysis results.Methods:According to the prescription analysis,chemical composition of medicinal materials,modern pharmacological mechanism and other comprehensive analysis,the feasibility of Deng’s Qingdu decoction in the prevention and control of COVID-19 was analyzed.The content of chlorogenic acid in Dengshi Qingdu liquid was determined by HPLC.The determination conditions were as follows:Chromatographic column:Agilent ZORBAX SB-C18(4.6 mm×250 mm,5-micron),mobile phase:acetonitrile-0.1%phosphoric acid(10:90),flow rate:1.0 mL/min,column temperature:25℃,detection wavelength:326 nm.Results:The comprehensive analysis of Deng's Qingdu Decoction from the aspects of prescription analysis,pharmacodynamic active ingredients and pharmacological mechanism could explain its anti-COVID-19 effect to a certain extent.The linear range of chlorogenic acid was 13.91-278.25μg/mL(r=0.9999),and the average recovery was 100.03%[relative standard deviation(RSD)=1.23%].Conclusion:Deng’s Qingdu Decoction can be used for the prevention and control of COVID-19.The content determination method established in this paper can be used to determine the content of chlorogenic acid in Deng’s Qingdu decoction.
