丁二烯暴露工人外周血淋巴细胞凋亡分析

Analysis on apoptosis of peripheral blood lymphocytes in workers exposed to butadiene

摘要 目的 研究丁二烯(BD)暴露工人外周血淋巴细胞凋亡状况,进一步探讨BD的健康损害机制。方法 收集42名BD暴露工人及56名非暴露人群的班末尿,超高效液相色谱-电轰击离子源-质谱联用方法测定尿中R,S-1,2-双羟基-4-(N-乙酰半胱氨酸)-丁烷(DHBMA)浓度,按其浓度检测结果将研究对象分为低、中、高BD暴露组。采用Annexin V/FITC-PI试剂盒双染后经流式细胞仪(FCM)测定淋巴细胞凋亡率。结果 暴露组淋巴细胞早期凋亡率为(7.63±2.04)%,高于对照组(6.91±1.61)%,但差异无统计学意义(P=0.066)。低、中、高BD暴露组淋巴细胞早期凋亡率分别为(7.08±1.67)%、(7.59±2.58)%和(8.18±1.82)%,各组间差异无统计学意义(P>0.05)。以对照组淋巴细胞凋亡率的10.1%为界值判断淋巴细胞凋亡阳性率,凋亡阳性率随BD暴露浓度的增高有所升高;相关分析亦显示,尿中DHBMA浓度与外周血淋巴细胞早期凋亡率呈正相关(Kendall’s r=0.153,P<0.05)。结论 BD可导致暴露工人外周血淋巴细胞凋亡率轻度升高,似与尿中DHBMA水平有一定相关。 Objective To study the situation of apoptosis in peripheral blood lymphocytes of butadiene(BD) exposed workers,and further discuss the injury mechanism of BD.Methods Urine samples from 42 BD exposed workers and 56 controls were collected after work,and the concentration of urinary DHBMA was determined by UPLC-ESI-MS/MS,the the BD exposed workers were divided into low,medium and high exposure groups according to the concentration of urinary DHBMA.The apoptosis rate of lymphocytes was measured by flow cytometry(Annexin V/FITC-PI double stained).Results The early apoptosis rate of lymphocytes in BD exposed group was(7.63±2.04)%,which was higher than that in the control group(6.91±1.61)%,but there was no statistics significance(P=0.066);the early apoptosis rates of lymphocytes in different BD exposure level groups were(7.08±1.67)%,(7.59±2.58)% and(8.18±1.82)%,respectively,the difference was also no statistics significance(P>0.05).If 10.1% lymphocyte apoptosis rate in the control group was used as the boundary value of the positive rate of lymphocyte apoptosis,the positive rate of lymphocytes apoptosis showed some rise with the increase of BD exposure concentration.Correlation analysis also showed that urine DHBMA concentration was also positively correlated with the early apoptosis rate of peripheral blood lymphocytes(Kendall’s r=0.153,P<0.05).Conclusion BD exposure can increase some lymphocyte apoptosis rate,which seems related to urinary DHBMA level.
