Determination of twenty-six inorganic elements in Polygonati odorati Rhizoma and Polygonati Rhizoma from different growing areas by ICP-MS

摘要 目的建立电感耦合等离子体-质谱(ICP-MS)法测定不同产地玉竹和黄精中Na、Mg、Al、K、Ca、Mn、Fe、Ni、Cu、Zn、V、Cr、Co、Ga、As、Se、Rb、Sr、Ag、Cd、Cs、Ba、Tl、Pb、U、Hg的含量。方法40批样品经微波消解后,采用ICP-MS法测定各无机元素含量,绘制其图谱,主成分分析(PCA)、正交偏最小二乘法判别分析(OPLS-DA)进行多变量统计分析。结果2种药材主要无机元素均为K、Mg、Ca、Al、Fe、Mn、Na,并存在正相关性。各无机元素峰形分布相似,其图谱具有特征性。不同产地2种药材可区分,Sr、Se、V、Cd、Al、Ga、Fe、Cr、Ni、Co是特征无机元素。Hg、Cr含量分别在5、6批样品中超标。结论该方法稳定可靠,可有效区分不同产地玉竹和黄精,并为其质量控制及安全评价提供依据。 AIM To establish an inductively coupled plasma-mass spectrometry(ICP-MS) method for the content determination of Na, Mg, Al, K, Ca, Mn, Fe, Ni, Cu, Zn, V, Cr, Co, Ga, As, Se, Rb, Sr, Ag, Cd, Cs, Ba, Tl, Pb, U and Hg in Polygonati odorati Rhizoma and Polygonati Rhizoma from different growing areas.METHODS Forty batches of samples were digested by microwave, after which ICP-MS was applied to determining the contents of various inorganic elements, whose spectra were drawn, principal component analysis(PCA) and orthogonal partial least squares discriminant model analysis(OPLS-DA) were used for multivariate statistical analysis.RESULTS The main inorganic elements in these two medicinal materials were K, Mg, Ca, Al, Fe, Mn and Na, which existed certain positive correlations. Various inorganic elements demonstrated similiar peak shape distribution, whose spectra had characteristics. Through the orthogonal partial least squares discriminant mode analysis(OPLS-DA) method, the difference in the content of inorganic elements in the two samples can be compared, Two medicinal materials from different grwoing areas can be distinguished, Sr, Se, V, Cd, Al, Ga, Fe, Cr, Ni and Co were characteristic inorganic elements. Hg and Cr contents exceeded standard in five and six batches of samples, respectively.CONCLUSION This stable and reliable method can be used for the effective distinguishment of Polygonati odorati Rhizoma and Polygonati Rhizoma from different growing areas, and provide references for their quality control and safety evaluation.
