Quality evaluation of Astragali Radix from different producing areas by HPLC-CAD fingerprints combined with chemometrics

摘要 目的 通过HPLC-CAD指纹图谱结合化学计量学评价黄芪质量。方法 分析采用Agilent SB-C色谱柱(150 mm×4.6 mm, 3.5μm);流动相乙腈-0.1%甲酸,梯度洗脱;体积流量1.5 mL/min;柱温40℃;蒸发光散射检测。利用中药色谱指纹图谱相似度评价系统结合聚类分析、主成分分析(PCA)及偏最小二乘法-判别分析(PLS-DA)对3个主要产地25批样品进行质量评价。结果 HPLC-CAD指纹图谱中有20个共有峰,鉴定出10个,相似度0.833~0.979。不同产地样品中化学成分存在差异,筛选出10个差异标志物。结论 该方法稳定可靠,可用于不同产地黄芪的质量控制和评价。 AIM To evaluate the quality of Astragali Radix from different producing areas by HPLC-CAD fingerprints combined with chemometrics.METHODS The analysis was conducted on a 40 ℃ thermostatic Agilent SB-Ccolumn(150 mm×4.6 mm, 3.5 μm), with the mobile phase comprising of acetonitrile-0.1% formic acid flowing at 1.5 mL/min in a gradient elution manner, and the detection was set by CAD. Twenty-five batches of samples from three different producing areas had their quality evaluated using “Similarity Evaluation System for Chromatographic Fingerprint of Chinese Materia” combined with cluster analysis, principal component analysis(PCA), and partial least squares-discriminant analysis(PLS-DA).RESULTS Among the twenty common peaks in the HPLC-CAD fingerprints, ten of whcih sharing similarities within the range of 0.833-0.979 were identified. Therefore, for the samples of different producing areas with chemical constituents, variations, ten identifiable difference markers were screened.CONCLUSION This stable and reliable method can be used for quality control and evaluation of Astragali Radix from different producing areas.
