菟丝子总黄酮对先兆流产大鼠妊娠结局及Th1/Th2平衡的作用机制研究 被引量:11

Effect of Total Flavonoids of Cuscuta chinensis on Pregnancy Outcome and Th1/Th2 Balance in Threatened Abortion Rats

摘要 目的探讨菟丝子总黄酮(TFCC)对先兆流产(TA)大鼠妊娠结局的影响及机制。方法妊娠大鼠随机分为对照组、模型组、TFCC低、高剂量组及黄体酮组,于妊娠第1天给药,10 d后,除对照组外,均构建TA大鼠模型。妊娠12 d后比较妊娠结局;HE染色观察子宫组织病理;ELISA法检测雌二醇、黄体酮、IL-4、IFN-γ、TNF-α及IL-10水平;RT-qPCR技术检测子宫GATA-3、IL-4、T-bet及IFN-γmRNA表达量;Western blot技术检测IL-4、IFN-γ蛋白表达量。结果与对照组比较,模型组孕鼠活胚数减少,死胚数及流产率升高(P<0.05),子宫内膜结构见有炎性损伤,血清雌二醇、黄体酮、IL-4、IL-10及子宫GATA-3 mRNA、IL-4 mRNA及蛋白相对表达量降低(P<0.05),血清IFN-γ、TNF-α及子宫T-bet mRNA、IFN-γmRNA及蛋白相对表达量升高(P<0.05);与模型组比较,各给药组孕鼠的上述指标均逆转;且TFCC低、高剂量及黄体酮的作用效果逐渐增强(P<0.05)。结论TFCC可改善TA大鼠妊娠结局,其作用机制可能与调节Th1/Th2细胞因子失衡有关。 Objective To investigate the effect and mechanism of total flavonoids of Cuscuta chinensis(TFCC)on pregnancy outcome in threatened abortion(TA)rats.Methods Pregnant rats were randomly divided into control group,model group,TFCC low and high dose group and progesterone group.They were administered on the first day of pregnancy.After 10 d,TA rat models were constructed except the control group.The pregnancy outcomes were compared after 12 d of pregnancy.HE staining was used to observe the histopathology of uterus.Estradiol,progesterone,IL-4,TNF-α,IL-10 and IFN-γlevels were detected by ELISA.Uterine GATA-3,IL-4,T-bet and IFN-γmRNA expression were detected by RT-qPCR.IL-4 and IFN-γprotein expression were detected by Western blot.Results Compared with the control group,the number of live embryos in the model group decreased,the number of dead embryos and abortion rate were increased,and inflammatory damage was found in the structure of endometrium,the relative expressions of serum estradiol,progesterone,IL-4,IL-10 and uterine GATA-3 mRNA,IL-4 mRNA and protein were decreased,serum IFN-γ,TNF-αand the relative expression of uterine T-bet,IFN-γmRNA and protein were increased(P<0.05).Compared with the model group,the above indexes of pregnant rats in the administration groups were reversed(P<0.05).The effiects of low and high doses of TFCC and progesterone were gradually enhanced(P<0.05).Conclusion TFCC can improve the pregnancy outcome of TA rats,and its mechanism may be related to the regulation of Th1/Th2 cytokine imbalance.
