温郁金对胃癌细胞的抑制作用及其对IGF-Ⅰ,IGF-Ⅱ表达的影响

Inhibitory effect of wenyujin on gastric cancer cells and its effect on the expression of IGF-Ⅰ and IGF-Ⅱ

目的:探讨温郁金对胃癌细胞的抑制作用及其对IGF-Ⅰ、IGF-Ⅱ表达的影响。方法:经超临界二氧化碳萃取法,提取温郁金挥发油成分,选择氟尿嘧啶(5-Fu)作为阳性对照物,采取MTT法,检测温郁金提取物对胃癌细胞的影响,通过放射免疫法观察IGF-Ⅰ、IGF-Ⅱ浓度变化。结果:(1)随着剂量增加,温郁金提取物1、2对胃癌细胞的抑制率呈增加趋势,具有统计学意义(P<0.05)。(2)提取物1、2的IGF-Ⅰ、IGF-Ⅱ含量明显低于对照组,差异存在统计学意义(P<0.05)。结论:温郁金提取物对胃癌细胞生长具有抑制作用,且抑制作用可能与IGF-Ⅰ、IGF-Ⅱ表达有关。

Objective:To investigate the inhibitory effect of wenyujin on gastric cancer cells and its effect on the expression of IGF-I and IGF-II.Methods:the volatile oil of Curcuma wenyujin was extracted by supercritical carbon dioxide extraction.Fluorouracil(5-FU)was selected as the positive control.The effect of Curcuma wenyujin extract on gastric cancer cells was detected by MTT method.The concentration changes of IGF-Ⅰand IGF-Ⅱwere observed by radioimmunoassay.Results:(1)with the increase of dose,the inhibitory rate of wenyujin extract 1 and 2 on gastric cancer cells increased,which was statistically significant(P<0.05).(2)The contents of IGF-I and IGF-II in extracts 1 and 2 were significantly lower than those in the control group(P<0.05).Conclusion:wenyujin extract has inhibitory effect on the growth of gastric cancer cells,and the inhibitory effect may be related to the expression of IGF-Ⅰand IGF-Ⅱ.
