箭筈豌豆种质资源的主要品质评价

摘要 [目的]为了筛选出优质的箭筈豌豆蛋白种质,对来自山东省农业科学院农作物种质资源研究所的123份箭筈豌豆种质资源的主要品质(氢氰酸含量、粗蛋白含量、可溶性蛋白含量、淀粉含量)进行评价。[方法]首先采用苦味酸分光光度法测定氢氰酸含量,根据测定结果,筛选出58份箭筈豌豆种质;再采用凯氏定氮法测定粗蛋白含量、考马斯亮蓝法测定可溶性蛋白含量、试剂盒法测定淀粉含量,并对其进行相关性分析。[结果]箭筈豌豆种质中氢氰酸含量与粗蛋白含量呈显著负相关,与淀粉含量呈显著正相关。以粗蛋白含量为主要评价依据,淀粉含量和可溶性蛋白含量作为次要评价依据,筛选出CV32和CV34作为优良种质资源,2份材料的粗蛋白和淀粉总含量都达到75%以上,并且可溶蛋白含量达到15%以上。[结论]该研究为箭筈豌豆品种的培养提供科学依据。 [Objective]To screen the high-quality Vicia sativa protein germplasm,the main quality of 123 Vicia sativa germplasm samples from the Crop Germplasm Resources Research Institute of Shandong Academy of Agricultural Sciences(hydrocyanic acid content,crude protein content,soluble protein content and starch content)were evaluated.[Method]At first,the content of hydrocyanic acid was determined by picric acid spectrophotometry,according to the results,58 Vicia sativa germplasm were screened out.Next,the content of crude protein was determined by Kjeldahl nitrogen determination method,soluble protein content was determined by Coomash bright blue method,starch content was determined by kit method,and its correlation was analyzed.[Result]The content of hydrocyanic acid in Vicia sativa germplasm was significantly negatively correlated with the content of crude protein,but significantly positively correlated with the content of starch.Taking the content of crude protein as the main screening basis,and the content of starch and soluble protein as the secondary screening basis,CV32 and CV34 were selected as excellent germplasm resources.The total content of crude protein and starch in these two materials reached more than 75%,and the content of soluble protein reached more than 15%.[Conclusion]This study provides a scientific basis for the cultivation of pea varieties with arrows.
