艾灸对D-半乳糖致亚急性衰老模型大鼠皮肤氧化应激及血清IL-6的影响

Effects of Moxibustion on Skin Oxidative Stress and Serum IL-6 in Subacute Aging Model Rats Induced by D-galactose

目的:观察艾灸对D-半乳糖致亚急性衰老模型大鼠皮肤组织氧化指标和血清IL-6的影响,探讨艾灸抗皮肤衰老作用及其机制。方法:采用D-半乳糖制备亚急性衰老模型,除正常组外,其余各组每日颈背部皮下注射10%D-半乳糖(1000 mg·kg^(-1)),正常组注射同等剂量的生理盐水,连续42 d。造模的第22天,艾灸组予以艾灸“关元”穴及双侧“足三里”穴,阳性对照组颈背部涂抹维生素E。42 d后,检测大鼠皮肤组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量以及血清白介素-6 (IL-6)的浓度。结果:与正常组比较,模型组大鼠皮肤组织中SOD活性显著降低,MDA含量明显升高,血清IL-6水平显著升高(P<0.01)。与模型组比较,艾灸可增强大鼠皮肤组织SOD活性,降低MDA含量,降低血清IL-6水平(P<0.01)。结论:艾灸可通过调节氧化应激紊乱及免疫因子来延缓皮肤衰老。

Objective To observe the effects of moxibustion on skin oxidation index and serum IL-6 in subacute aging model rats induced by D-galactose, and to explore the anti-skin aging effect and its mechanism.Methods Subacute aging model was established by D-galactose. Except for the normal group, the other groups are subcutaneously injected with 10% D-galactose(1000 mg·kg^(-1)) daily. The normal group was injected with the same dose of saline for consecutive 42 days. On the 22th day of modeling, the moxibustion group was treated with moxibustion “Guan yuan” point and bilateral “Zu San li” point, and while Vitamin E cream was smeared on the neck and back of positive control group. After 42 days, the activity of superoxide dismutase(SOD), the content of malondialdehyde(MDA) and the concentration of interleukin-6(IL-6) in serum were detected.Results Compared with the normal group, SOD activity in skin tissue of model group was significantly decreased, MDA content was significantly increased, and serum IL-6 level was significantly increased(P< 0. 01). Compared with the model group, moxibustion increased SOD activity, decreased MDA content, and decreased serum IL-6 level(P< 0. 01).Conclusion Moxibustion can delay skin aging by regulating oxidative stress disorder and immune factors.
