妊娠期糖尿病胎盘组织中SOCS3与IRS1、IRS1^(ser307)水平的相关性研究

Correlation of SOCS3 and IRS1 and IRS1^(ser307) Levels in Placental Tissue of Gestational Diabetes

摘要 目的:探索妊娠期糖尿病(GDM)胎盘组织中SOCS3与IRS1、IRS1^(ser307)水平的相关性。方法:收集GDM组及正常孕妇组胎盘组织。HE染色观察GDM胎盘组织的病理变化。QRT-PCR、免疫组化检测SOCS3、IRS1在胎盘组织中的表达及分布情况。Western blot检测SOCS3、IRS1、IRS1^(ser307)三种蛋白的表达量。Spearman秩和相关性分析胎盘组织中SOCS3与IRS1、IRS1^(ser307)蛋白表达的相关性。结果:(1)GDM胎盘组织胎盘血管分布紊乱,管壁增厚且管腔狭窄,血细胞减少。(2)SOCS3和IRS1主要特异性表达于细胞质中,SOCS3在GDM组中表达升高,呈黄色,为阳性表达;IRS1在GDM组中表达降低,几乎不表达。(3)QRT-PCR发现,与对照组相比,在GDM组胎盘中SOCS3表达上调(P<0.01);IRS1表达下调(P<0.01)。(4)Western Blot发现,与对照组相比,在GDM组胎盘中SOCS3表达上调(P<0.01);IRS1表达下调(P<0.01);IRS1^(ser307)表达上调(P<0.01)。(5)经分析发现SOCS3与IRS1之间的相关性(r=-0.635 4,P<0.000 1);SOCS3与IRS1^(ser307)之间的相关性(r=0.902 8,P<0.000 1)均呈强相关。结论:SOCS3和IRS1^(ser307)在GDM胎盘组织中表达上调,IRS1表达下调。SOCS3与IRS1呈负相关,与IRS1^(ser307)呈正相关。 Objective:To explore the correlation between SOCS3 in placental tissue of Gestational diabetes mellitus(GDM) and the levels of IRS1 and IRS1^(ser307).Methods:Placental tissues were collected in the GDM women(GDM group) and in the normal pregnant women(control group).The pathological changes of GDM placenta tissue were observed by HE staining.The expression and distribution of SOCS3 and IRS1 in placental tissues were detected by QRT-PCR and immunohistochemistry.The expressions of SOCS3,IRS1 and IRS1^(ser307) were detected by Western blot.Spearman rank sum correlation analysis of the correlation between SOCS3 and IRS1,IRS1^(ser307) protein expression in placental tissues.Results:(1)GDM placental tissue had disordered placental blood vessel distribution, thickened tube wall and narrowed lumen, and decreased blood cells.(2)SOCS3 and IRS1 were mainly expressed specifically in the cytoplasm.SOCS3 were elevated in the GDM group, which was yellowish and positive expression;IRS1 was reduced in the GDM group and almost not expressed.(3)QRT-PCR found that compared with the control group, the expression of SOCS3 was up-regulated in GDM group placenta(P<0.01);the expression of IRS1 was down-regulated(P<0.01).(4)Western Blot found that compared with the control group, the expression of SOCS3 in GDM group placenta was up-regulated(P<0.01);the expression of IRS1 was down-regulated(P<0.01);the expression of IRS1^(ser307) was up-regulated(P<0.01).(5)After analysis, it was found that the correlation between SOCS3 and IRS1(r=-0.635 4,P<0.000 1);the correlation between SOCS3 and IRS1^(ser307)(r=0.902 8,P<0.000 1),and both showed strong correlation.Conclusion:SOCS3 and IRS1^(ser307) were up-regulated in GDM placental tissues, and IRS1 was down-regulated.SOCS3 was negatively correlated with IRS1,and positively correlated with IRS1^(ser307).
