摘要 Hedgehog(Hh)信号通路在脊椎动物的各种生理过程中都发挥着重要作用,包括细胞的生长和分化等。Sonic hedgehog(Shh)信号通路是Hh信号通路中研究最深入的成员之一,主要在真核生物中发挥生物学功能。国内外多项研究表明,人类各种肿瘤的发生发展都涉及Shh信号通路,多种因素引起的Shh异常活化会导致人体系统多种恶性肿瘤的发生。神经胶质瘤相关癌基因同源蛋白(glioma-associated oncogene homologue,Gli)是Shh信号通路的关键成员之一,参与调控肿瘤的发生和进展。该文总结了Shh信号通路中Gli蛋白激活的分子机制及其在肿瘤发生发展中的作用,并讨论了Gli蛋白与肿瘤干细胞的相互作用,以及Gli蛋白靶向治疗相关肿瘤的新思路。 Hedgehog(Hh)signaling pathway plays an significant role in a variety of physiological processes in vertebrates,including cell growth and differentiation.The Sonic hedgehog(Shh)signaling pathway is one of the most intensively studied members of the Hh signaling pathway,which plays a biological function in eukaryotes.A considerable number of research results at home and abroad have shown that the participation of the Shh signaling pathway is involved in the occurrence and development of various human tumors.Abnormal activation of Shh induced by various factors can lead to the occurrence of various malignant tumors of the human body systems.Glioma-associated oncogene homologue(Gli)is one of the key members of the Shh signaling pathway,which regulates the occurrence and progression of tumors.This article summarizes the molecular mechanism of Gli protein activation in the Shh signaling pathway and its role in tumorigenesis and development.At the same time,it also discusses the interaction between Gli protein and cancer stem cells and new ideas in Gli protein targeted therapy for related tumors.
