外周血浆3种微小RNA水平与2型糖尿病及微血管病变的临床研究 被引量:1

Clinical trial of the levels of 3 kinds of microRNAs in peripheral blood plasma and type 2 diabetes mellitus combined with microvascular complications

摘要 目的 探讨2型糖尿病(type 2 diabetes mellitus,T2DM)及微血管病变患者血浆中miR-146a、miR-155和miR-181a的变化。方法 选取44例T2DM患者为研究对象,其中单纯T2DM患者23例(对照组),T2DM合并微血管病变21例(试验组),并以同期健康体检人群44例作为正常组。用实时荧光定量聚合酶链反应检测3组受试者血浆中miR-146a、miR-155和miR-181a的表达水平。结果 校正性别、年龄和生化指标之后,对照组和试验组外周血血浆中miR-146a、miR-155、miR-181a表达量(0.55±0.17和0.62±0.17,0.61±0.23和0.70±0.41,0.91±0.28和0.97±0.42)均明显低于正常组(1.16±1.30,1.12±1.15,1.52±1.29),差异均有统计学意义(均P<0.05),而对照组和试验组的3种miRNAs表达量差异均无统计学意义(均P>0.05)。结论 T2DM及微血管病变患者血浆中miR-146a、miR-155和miR-181a表达量均明显下调,其可能参与了T2DM及微血管病变的发生发展。 Objective To explore the relationship between the expression levels of miR-146a,miR-155 and miR-181a in plasma and type 2 diabetes mellitus(T2 DM) combined with microvascular complications.Methods A total of 44 T2 DM patients were selected as the research objects,including 23 cases of T2 DM patients alone(control group) and 21 cases of T2 DM complicated with microvascular disease(treatment group),and 44 healthy physical examination population during the same period were regarded as the normal group.The expression levels of miR-146a,miR-155 and miR-181a were detected by real-time polymerase chain reaction.Results After adjusting the age,sex and biochemical indicators,the expression levels of miR-146a,miR-155 and miR-181a in control and treatment groups(0.55±0.17 and 0.62±0.17,0.61±0.23 and 0.70±0.41,0.91±0.28 and 0.97±0.42,respectively) were lower than that of in normal group(1.16±1.30,1.12±1.15,1.52±1.29,respectively),and the differences were statistically significant(all P<0.05),while the expression levels of three miRNAs between the control group and treatment group were not statistically significant(all P>0.05).Conclusion The plasma expression levels of miR-146a,miR-155 and miR-181awere down-regulated in the patients of T2DM combined with microvascular complications,suggesting thatthey may be involved in the occurrence and development of T2DM combined with microvascular complications.
