从“脾主运化”探讨半夏泻心汤对PCOS大鼠肠道菌群以及糖脂代谢的影响 被引量:1

Effects of Banxia Xiexin Decoction on intestinal microflora and glucolipid metabolism in PCOS rats

摘要 目的探讨半夏泻心汤对PCOS模型大鼠血清性激素水平和肠道菌群以及糖脂代谢的影响。方法将SD大鼠随机分成对照组、模型组(来曲唑1 mg·kg^(-1)·d^(-1))、达英-35组(0.18 mg·kg^(-1)·d^(-1))、二甲双胍组(0.135 g·kg^(-1)·d^(-1))、半夏泻心汤组(5.31 g·kg^(-1)·d^(-1))以及半夏泻心汤+二甲双胍+达英-35(3种药物合用)组,每组10只,灌胃28天,检测大鼠体重,用ELISA法检测血清性激素水平、血糖(FBG)、胰岛素指数(HOMA-IR)、总胆固醇(TG)和甘油三酯(TC)变化以及用16s rDNA扩增子焦磷酸测序肠道菌群变化。结果半夏泻心汤明显减低PCOS模型大鼠体重,血清促黄体生成素(LH)和睾酮(T)水平,HOMA-IR、TG和TC水平表达,增加肠道菌数量、多样性以及菌群均匀度,门水平增加拟杆菌门(Bacteroidetes)、变形菌门(Proteobacteria)、放线菌门(Actinobacteria)、疣微菌门(Verrucomicrobia)相对丰度,降低厚壁菌门(Firmicutes);在属水平上增加有益菌:拟杆菌属(Bacteroides)、萨特氏菌属(Sutterella)、乳酸菌属(Lactoba⁃cillus)、双歧杆菌属(Bifidobacterium)和艾克曼菌属(Akkermansia muciniphila)相对丰度,而降低致病菌:普氏菌属(Prevotel⁃la)、瘤胃球菌属(Ruminococcus)、颤螺菌属(Oscillospira)和厌氧菌属(Anaeroplasma)相对丰度。结论半夏泻心汤降低P⁃COS大鼠血清高雄激素和高胰岛素血症的作用机制,可能与提高肠道有益菌相对丰度、降低致病菌相对丰度有关。 Objective To investigate the effects of Banxia Xiexin Decoction on serum sex hormone levels,intestinal flora and glu⁃cose and lipid metabolism in PCOS model rats.Methods SD rats were randomly divided into control group,model group(Letro⁃zole 1 mg·kg^(-1)·d^(-1)),dae-35 group(0.18 mg·kg^(-1)·d^(-1)),metformin group(0.135 g·kg^(-1)·d^(-1))and BXD group(5.31)g·kg^(-1)·d^(-1)and BXD+metformin+daca-35(3 drugs combined),10 rats in each group,and gavage for 28 days.Body weight was detected,serum sex hormone levels,blood glucose(FBG),insulin index(HOMA-IR),total cholesterol(TG)and triglyceride(TC)were determined by ELISA,and intestinal microflora was sequenced by pyrosequencing with 16S rD⁃NA amplification.Results Banxia Xiexin Decoction significantly reduced the body weight,serum luteinizing hormone(LH)and testosterone(T)levels,and the expressions of HOMA-IR,TG and TC levels in PCOS model rats,and increased the number,diversity and evenness of intestinal bacteria.Phylum level increased the relative abundance of Bacteroidetes,Proteobacteria,Acti⁃nobacteria and Verrucomicrobia,while decreased Firmicutes.To add beneficial bacteria at the genus level:Relative abundance of Bacteroides,Sutterella,Lactobacillus,Bifidobacterium and Akkermansia muciniphila,The relative abundance of Prevotella,Rumi⁃nococcus,Oscillospira and Anaeroplasma was decreased.Conclusion The mechanism of Banxia Xiexin Decoction reducing serum hyperandrogen and hyperinsulinemia in PCOS rats may be related to increasing the relative abundance of intestinal beneficial bac⁃teria and decreasing the relative abundance of pathogenic bacteria.
