马钱苷对神经系统相关疾病的生物活性研究

Study on bioactivities of loganin on nervous system related diseases

摘要 马钱苷是山茱萸所含的一个重要的药效成分和质控成分,具有广泛的生物活性。文章通过对中国知网(CNKI)、PubMed等国内外常用的专业数据库的检索分析,总结梳理了近年来关于山茱萸马钱苷对神经系统相关疾病的生物活性研究。研究结果表明,山茱萸马钱苷具有抗抑郁、改善学习记忆障碍、缓解神经性疼痛、缓解帕金森病、改善阿尔兹海默病、镇静、催眠、改善糖尿病神经病变、改善脊髓性肌萎缩症、缓解糖尿病并发抑郁症、缓解糖尿病并发焦虑症、抗脑缺血再灌注损伤、抗雪旺氏细胞焦亡等中枢神经系统疾病广泛的生物活性。以其抗抑郁研究为代表进行总结梳理显示,马钱苷(12.5,50 mg/kg)给药3 d对雄性小鼠悬尾实验测试呈现出抗抑郁作用,给药10 d对链脲佐菌素诱导的糖尿病雄性大鼠的抑郁样行为具有改善作用,其潜在的抗抑郁机制涉及调控5-羟色胺能神经系统、降糖以及抑制炎症等有关,并据当前研究现状提出建议:开展基于更长时程给药的抗抑郁药效及其量效关系研究、在炎症模型上考察马钱苷的抗抑郁样的作用及其机制、开展马钱苷抗抑郁作用的性别差异研究、以及开展马钱苷抗抑郁确切的分子靶点研究。该研究为山茱萸及其主要药效物质马钱苷在防治中枢神经系统相关疾病的进一步研发应用提供了文献支撑和思路启发。 Loganin is an important medicinal component and quality control component contained in Cornus officinalis,and it has a wide range of biological activities.The article summarizes and combs the research on the biological activity of C.officinalis on nervous system-related diseases in recent years by searching and analyzing the professional databases commonly used at home and abroad such as China Knowledge Network(CNKI)and PubMed.The results of the study show that loganin has anti-de⁃pressant,improve learning and memory impairment,relieving neuropathic pain,relieving Parkinson’s disease,improving Alzhei⁃mer’s disease,sedation,hypnosis,improving diabetic neuropathy,improving spinal muscular atrophy,relieving diabetes complicated with depression,relieving diabetes complicated with anxiety,anti-cerebral ischemia-reperfusion injury,anti-Schwann's cell pyrolysis with a wide range of biological activities,especially anti-depressive research is more active.Taking its antidepressant research as a representative,a summary and combing showed that loganin(12.5,50 mg/kg)administration for 3 days showed antidepressant effect on male mice tail suspension test,and administration for 10 days had an ameliorating effect on strep⁃tozotocin-induced depression-like behaviors of diabetic male rats,and its potential antidepressant mechanism involves regula⁃ting the serotonergic nervous system,reducing blood sugar,and inhibiting inflammation.According to the current research status,suggestions are made:Study on the antidepressant effect of time-course administration and its dose-effect relationship,investi⁃gate the antidepressant-like effect and mechanism of loganin on an inflammation model,conduct gender differences in loganin's antidepressant effect,and study on the exact molecular targets of loganin for antidepressant.This study provides literature support and inspiration for the further development and application of C.officinalis and its main medicinal substance loganin in the prevention and treatment of central nervous system-related diseases.
