HPLC法测定不同产地当归处理中7种成分的含量 被引量:8

HPLC analysis and study of the content determination of 7 components of Angelica sinen⁃sis in four producing areas in different treatments

摘要 目的建立HPLC法测定当归根中7种成分含量的方法,对比分析评价12批来自不同产地及同产地不同处理当归的质量。方法采用ZORBAX SB-C_(18)色谱柱(250 mm×4.6 mm,5μm),以1%冰乙酸水(A)-乙腈(B)为流动相梯度洗脱,流速1.0 mL/min,检测波长280 nm,柱温30℃,进样量10μL。并用聚类、主成分分析对当归不同处理样品质量进行比较研究。结果当归根中7种成分分离度良好,12批不同产地处理的当归根中7种成分含量有较大差异。P值分析、聚类分析及主成分分析结果表明,当归根中7种成分在不同产地以阿魏酸松柏酯、Z-藁本内酯差异大,且聚为3类,能显著分开,当归产地以宕昌八力镇中拉村的质量相对最优。结论所建立的方法简便准确,重复性好,岷县、宕昌等地当归质量优于天祝、榆中等地,但通过施用合理处理能提高当归质量来缩小差异。 Objective To establish an HPLC method to determine the contents of 7 components in the roots of Angelica sinensis,and to compare and analyze the quality of 12 batches of Angelica sinensis from different producing areas and different treatments in the same producing area.Methods A ZORBAX SB-C_(18) column(250 mm×4.6 mm,5μm)was used,with 1%glacial acetic acid water(A)-acetonitrile(B)as mobile phase gradient elution,flow rate 1.0 mL/min,detection wavelength 280 nm,the column tem⁃perature was 30℃,and the injection volume was 10μL.Clustering and principal component analysis were used to compare the quality of different treatment samples of Angelica sinensis.Results The separation of 7 components in the roots of Angelica sinensis keiskei koidz was good,and the contents of 7 components in the roots of the 12 batches of Angelica sinensis keiskei treated from different producing areas were quite different.The results of P value analysis,cluster analysis and principal component analysis showed that the 7 components of Angelica sinensis root are very different in different producing areas with coniferyl ferulate and Z-ligustilide,and they are grouped into 3 categories,which can be significantly separated.The place of production is relatively best in Zhongla Village,Bali Town,Tanchang.Conclusion The established method is simple,accurate and reproducible.The quality of Angelica sinensis in Minxian and Tanchang is better than that in Tianzhu and Yuzhong,but the quality of Angelica sinensis can be improved by applying reasonable treatment to reduce the difference.
