摘要
目的:研究CALCR基因和自噬相关基因在非小细胞肺癌(Non-small-cell lung cancer,NSCLC)中的表达及相关性。方法:2021年9月到2022年1月共收集20例NSCLC标本,利用对应的癌旁组织作为对照。采用免疫印迹方法检测各组肺组织中CALCR、Beclin-1、ATG5、ATG13及p53蛋白表达情况,并采用SPSS统计软件进行病理因素及相关性分析。结果:与癌旁组织比较,NSCLC组中的CALCR蛋白表达明显上调(P<0.001),而自噬相关蛋白Beclin-1、ATG5、ATG13及p53明显下调(P<0.05);且CALCR蛋白水平在临床特征如性别,年龄,肿瘤部位及大小组间无显著性差异(P>0.05)。相关性分析显示CALCR与ATG5蛋白表达水平无相关性(r=0.189,P=0.6),而CALCR蛋白表达水平与ATG13(r=0.71,P=0.021),Beclin-1(r=0.63,P=0.038)及p53(r=0.655,P=0.04)蛋白表达水平呈显著正相关。结论:CALCR在NSCLC中高表达,而Beclin-1、ATG5、ATG13及p53在NSCLC中低表达;且CALCR与患者的临床特征无相关性,CALCR与ATG13、Beclin-1及p53呈正相关。
Objective:To study the expressions and correlation of CALCR gene and autophagy-related gene in non-small cell lung cancer(non-small-cell lung cancer,NSCLC).Methods:Specimens were taken from 20 patients admitted to our hospital from Sep 2021 to Jan 2022 with NSCLC and 20 adjacent tissues as control.The expressions of CALCR,beclin-1,ATG5,ATG13 and p53 proteins in lung tissues of each group were detected by Western blot,and the pathological factors and correlation were analyzed by SPSS statistical software.Results:compared with adjacent tissues,the protein expressions of CALCR in NSCLC group was significantly up-regulated(P<0.001),while autophagy related-proteins Beclin-1,ATG5,ATG13 and p53 were significantly down-regulated(P<0.05).No significant difference was observed in CALCR protein expression between gender,age,tumor location,tumor size and other groups(P>0.05).Correlation analysis showed that there was no correlation between CALCR and ATG5 protein expression level(r=0.189,P=0.6),but a significant correlation between CALCR protein expression level and ATG13(r=0.71,P=0.021),Beclin-1(r=0.63,P=0.038)and p53(r=0.655,P=0.04).Conclusion:CALCR protein was highly expressed in NSCLC,while Beclin-1,ATG5,ATG13 and p53 protein were low expressed in NSCLC.CALCR protein was not related to clinical features.The expression level of CALCR protein was significantly positively correlated with the expression levels of ATG13,Beclin-1 and p53 protein.
