刃天青微孔板法在本科实验教学中的应用

摘要 刃天青微孔板法是一种新近开发的快速测定抑菌活性的方法,已广泛应用于杀菌剂的高通量筛选及活性评价。文章以代表性病原真菌灰霉病菌为供试菌,应用刃天青微孔板法,设计了嘧菌酯杀菌剂对灰霉病菌的抑菌活性测定实验。与传统菌丝生长速率法相比,该方法具有样品消耗少、观察便捷、反应灵敏等优点,可用于杀菌剂活性定量测定实验,也可与农药化学、天然产物化学的杀菌剂合成或提取分离相关实验相结合,设计探究式实验,更好地培养学生的科研创新思维和综合实践能力,提高教学质量。 As a newly developed method for rapid determination of antimicrobial activity, resazurin microtitre plate assay has been widely used in high-throughput screening and activity evaluation of fungicides. Taking the representative pathogenic fungus Botrytis cinerea as the test strain, the antifungal activity of azoxystrobin fungicide against Botrytis cinerea was designed based on the resazurin microtitre plate assay. Compared with the traditional mycelial growth rate method, this method has the advantages of less sample consumption, convenient observation and sensitive response. It can be used for quantitative determination of fungicide activity, and can also be combined with fungicide synthesis or extraction and separation related experiments of pesticide chemistry and natural product chemistry to design exploratory experiments, so as to better cultivate students’ scientific research innovation thinking and comprehensive practical ability, and improve teaching quality.
