摘要
为进一步研究番鸭羽色性状的遗传机制,运用Illumina高通量测序技术对黑羽及白羽番鸭皮肤组织进行转录组测序(RNA-seq),利用实时荧光定量PCR(RT-qPCR)对部分RNA-seq数据进行验证后,经基因本体(GO)富集分析、京都基因与基因组百科全书(KEGG)通路注释以及蛋白互作网络数据库(STRING)对差异表达基因功能进行筛选和功能分析。结果显示:RT-qPCR验证结果与RNA-seq结果比对,差异表达基因变化趋势一致,黑羽番鸭与白羽番鸭皮肤中发现显著差异表达基因684个,其中上调差异表达基因362个,下调差异表达基因322个,进一步筛选获得番鸭羽色性状相关候选差异表达基因20个,其中溶质载体家族24成员4(SLC24A4)、SLC24A5、酪氨酸酶相关蛋白1(TYRP1)、多巴色素异构酶(DCT)、溶质载体家族7成员11(SLC7A11)和小眼畸形相关转录因子(MITF)可能对番鸭羽色的遗传调控起重要调控作用。试验结果丰富了番鸭羽色性状遗传机制的研究,并为番鸭的进一步选育提供了新的数据支撑。
In order to further understand the genetic mechanism of plumage color traits in Muscovy ducks,the transcriptome sequencing of skin tissues of black and white Muscovy ducks were performed by the Illumina high throughput sequencing technique.After RNA-seq data were validated by RT-qPCR,the functions of differentially expressed genes were screened and analyzed by GO enrichment analysis,KEGG pathway annotation and STRING of protein interaction.The results showed that there were 684 differentially expressed genes between the black Muscovy duck skin and the white Muscovy duck skin,of which 362 were up-regulated and 322 were down-regulated.20 candidate differentially expressed genes related to Muscovy duck feather color traits were further screened.Moreover,SLC24A4,TYRP1,DCT,SLC7A11,SLC24A5 and MITF might play an important role in the genetic regulation of black or white plumage traits in Muscovy ducks.These results enriched researches on the genetic mechanism of plumage color traits in Muscovy ducks,and provided new data support for further breeding of Muscovy ducks.
