Effect of ubiquitin-conjugating enzyme E2T on the proliferation,migration and invasion of ovarian cancer cells

目的:探究沉默泛素结合酶E2T(UBE2T)对卵巢癌细胞增殖、迁移及侵袭的影响。方法:实时荧光定量PCR检测UBE2T在人正常卵巢细胞系IOSE80和人卵巢癌细胞系HO8910和A2780中的表达;通过小干扰RNA技术对卵巢癌细胞进行si-UBE2T转染,Western blot检测转染效率。CCK8实验检测UBE2T对卵巢癌细胞增殖能力的影响;划痕愈合实验以及Transwell实验检测UBE2T对卵巢癌细胞迁移和侵袭的影响。结果:与IOSE80相比,HO8910和A2780细胞中UBE2TmRNA表达均升高(P<0.001);转染后,与对照组相比,si-UBE2T组的蛋白表达降低,细胞的增殖、迁移及侵袭能力也降低(P<0.05)。结论:沉默UBE2T能够抑制卵巢癌细胞增殖、迁移以及侵袭,UBE2T可能是卵巢癌治疗的潜在研究靶点。

Objective:To observe the effects of silent ubiquitin-conjugating enzyme E2T(UBE2T)on the proliferation,migration and invasion of ovarian cancer cells.Methods:Real-time fluorescence quantitative PCR was used to detect the expression of UBE2T in normal human ovarian cell line IOSE80 and human ovarian cancer cell lines HO8910 and A2780;Ovarian cancer cells were transfected with si-UBE2T by small interfering RNA technology,and then divided into control group and si-UBE2T group.Western blot assay was performed to measure the transfection efficiency.CCK8 experiment was conducted to examine the effect of UBE2T on the proliferation of ovarian cancer cells,and scratch healing and Transwell experiment were used to observe the effect of UBE2T on the migration and invasion of ovarian cancer cells.Results:Compared with IOSE80,UBE2TmRNA expression was increased in HO8910 and A2780 cells(P<0.001);After transfection,the protein expression was significantly reduced,and the proliferation,migration and cell invasiveness were remarkably reduced in the si-UBE2T group compared to the control group(all P<0.05).Conclusion:Silent UBE2T can inhibit the proliferation,migration and invasion of ovarian cancer cells,and may be a potential research target for treatment of ovarian cancer.
