角蛋白17 对大鼠胰腺纤维化模型的影响

摘要 目的:探讨角蛋白17(KRT17)对大鼠胰腺纤维化模型的影响。方法:健康SD雄性大鼠20只,随机分为对照组、模型组,其中模型组又分为4组(n=4)。模型组大鼠腹腔内注射10%的二乙基二硫代氨基甲酸盐(DDC)溶液,隔日1次,对照组大鼠腹腔内注射同等体积生理盐水。分别于实验后第7、15、22、30天,腹腔内注射麻药并处死各组大鼠。颈动脉采血,留取胰腺组织。用免疫组化法测定胰腺组织中Ⅰ型胶原表达水平;采用Western blot法检测胰腺组织中KRT17蛋白的表达;并用血细胞分析仪检测白细胞的含量,用分立式自动生化分析仪检测血浆中生化指标。结果:第15、22、30天组胰腺组织中I型胶原表达水平高于对照组(P<0.05);第22、30天组胰腺组织中I型胶原表达水平高于第7、15天组(P<0.05)。第15、22、30天组胰腺组织中白细胞计数、淋巴细胞绝对值、葡萄糖含量高于对照组(P<0.05);第22、30天组胰腺组织中白细胞计数、淋巴细胞绝对值、葡萄糖含量高于第7天组(P<0.05);第22天组胰腺组织中葡萄糖含量高于第15天组(P<0.05);第30天组胰腺组织中白细胞计数、淋巴细胞绝对值、葡萄糖含量高于第15天组(P<0.05)。第15、22、30天组胰腺组织中KRT17蛋白含量高于对照组(P<0.05);第22、30天组高于第7、15天组(P<0.05)。结论:在腹腔注射DDC溶液诱导的大鼠胰腺纤维化模型中,随着胰腺组织纤维化程度的加深,KRT17表达水平升高。 Objective:To investigate the effect of keratin 17(KRT17)on model rats with pancreatic fibrosis.Methods:Twenty healthy male SD rats were equally randomized to control group and model group.Then the rats in model group were further divided into 4 groups(n=4 for each group),and intraperitoneally administered with 10%DDC solution once every other day.Rats in the control group were intraperitoneally injected with the same volume of normal saline.At day 7,15,22,and 30 after experiment,anesthetic was injected into the abdominal cavity to sacrifice the rats in each group.Blood was taken via the carotid artery,and the pancreatic tissues were maintained.Expression level of collagen I and KRT17 protein in the pancreatic tissues were measured by immunohistochemistry and Western blot,respectively.Blood cell analyzer and discrete automatic biochemical analyzer were used to respectively detect the content of white blood cells and biochemical indexes in plasma.Results:The expression level of collagen I in the pancreatic tissue of the 15-day,22-day and 30-day group was significantly higher than that in the control group(P<0.05).Collagen I was highly expressed in the pancreatic tissues of the 22-day and 30-day group compared to the 7-day and 15-day group(P<0.05).The white blood cell count,absolute lymphocyte value,and glucose content in the pancreatic tissues of the 15-day,22-day and 30-day group were markedly higher than those in the control group(P<0.05),and were also notably higher in the 22-day and 30-day group than in the 7-day group(P<0.05).The glucose content in the pancreatic tissue in the 22-day group was significantly higher than that in the 15-day group(P<0.05),and the white blood cell count,absolute value of lymphocytes and glucose content in the pancreatic tissues were significantly higher in the 30-day group than in the 15-day group(P<0.05).KRT17 protein content in the pancreatic tissue was markedly higher in the 15-day,22-day and 30-day group than in the control group(P<0.05),and higher in the 22-day and 30-day group than in the 7-day and 15-day group(P<0.05).Conclusion:KRT17 expression increases with the severity of fibrosis of pancreatic tissues in the models rats induced by intraperitoneal injection of DDC solution.
