摘要
目的探讨维生素B_(6)(VB_(6))对非小细胞肺癌(NSCLC)的放疗增效作用。方法采用人NSCLC A549细胞进行体外低密度培养,用不同浓度的VB_(6)处理细胞,部分细胞在药物处理1天后用不同剂量的X线照射,计算克隆形成效率及存活分数。通过相互作用系数(CDI)、辐射增强比值(RER)评估VB_(6)增敏作用。结果VB_(6)呈剂量依赖性地抑制A549细胞克隆形成,随着VB_(6)浓度升高,细胞克隆数逐渐减少,克隆形成效率及存活分数均逐渐降低(P﹤0.05)。VB_(6)与X线联合处理细胞,可使X线照射对细胞存活的抑制作用更为明显(P﹤0.01)。VB_(6)与不同剂量(2、4、6 Gy)X线的CDI均小于1,RER均大于1。结论VB_(6)可抑制NSCLC细胞的存活,并且与X线联合应用具有辐射增敏及协同抑制效应,VB_(6)的这些特性使其有可能成为NSCLC放疗中联合治疗的候选制剂。
Objective To investigate the synergistic effect of vitamin B_(6)(VB_(6)) on radiotherapy for non-small cell lung cancer(NSCLC).Method Human NSCLC A549 cells were cultured at low density in vitro.Cells were treated with different concentrations of VB_(6).Among them,some cells were irradiated with different doses of X-rays after 1 day of drug treatment.The colony formation efficiency and survival fraction were calculated.VB_(6) sensitization was assessed by the radiation enhancement ratio(RER)and the coefficient of drug interaction(CDI).Result VB_(6) inhibited the colony for-mation of A549 cells in a dose-dependent manner.The number of cell clones decreased gradually,and the colony forma-tion efficiency and survival fraction decreased gradually with the increase of VB_(6) concentration(P<0.05).If VB_(6) and X ray were combined to treat cells,the inhibitory effect of X-ray irradiation on cell survival was more obvious(P<0.01).The CDI of VB_(6)and different doses(2,4,6 Gy)of X-rays were all less than 1,and the RER were all greater than 1.Con-clusion VB_(6) could inhibit the survival of NSCLC cells,and command with radiosensitization and synergistic inhibitory effects when combined with X-rays.These properties of VB_(6) mean that this target could be a potential candidate for com-bined therapy in NSCLC radiotherapy.
