维生素D_(3)对紫杉醇诱发大鼠周围神经病理性疼痛的影响 被引量:1

Effects of vitamin D_(3)on paclitaxel-induced peripheral neuropathic pain in rats

摘要 目的观察维生素D_(3)对紫杉醇(PTX)诱发的大鼠周围神经病理性疼痛的影响。方法将35只6周龄雄性SD大鼠按随机数字表法分为5组(n=7):对照组(Con组)、PTX溶剂组(PTX Vehicle组)、PTX组、PTX+维生素D_(3)溶剂组(PTX+VitD_(3)Vehicle组)和PTX+维生素D_(3)组(PTX+Vit D_(3)组)。分别在给药前(d 0)和给药后4、8、12、16及20 d测定5组大鼠50%右足底机械缩足反应阈值(PWTs)和热缩足潜伏期(TWL),并在d0,12及20 d测定5组大鼠血清25-羟基维生素D_(3)[25-(OH)D_(3)]水平。结果5组大鼠体质量在给药前(d0)及给药后差异均无统计学意义(均P>0.05)。与模型组相比,PTX+Vit D3组大鼠50%PWTs在8、12、16及20d时显著升高(均P<0.05),TWL在4、8、12、16及20d时显著升高(均P<0.01),血清25-(OH)D_(3)水平在12及20 d时显著升高(均P<0.01)。结论腹腔注射维生素D3能够在一定程度上缓解PTX诱发的大鼠周围神经病理性疼痛,为临床化疗痛的防治提供了新的探索方向。 Objective To observe the effect of vitamin D_(3)on paclitaxel-induced peripheralneuropathic painin rats.Methods Thirty-five 6-week-old male SD rats were divided into five groups according to the random number table method:control group(Con group),paclitaxel vehicle group(PTX vehicle group),paclitaxel group(PTX group),paclitaxel+vitaminD_(3)vehicle group(PTX+VitD_(3)vehicle group),paclitaxel+vitaminD_(3)group(PTX+VitD_(3)group),with seven rats in each group.The 50%paw withdrawal thresholds(PWTs)and thermal withdrawal latency(TWL)were measured before(d0)and d4,d8,d12,d16 and d20 after administration of the drug.Serum 25-hydroxyvitamin D_(3)[25-(OH)D_(3)]levels of five groups of rats were measured at d0,d12 and d20.Results There was no significant difference in body weight among five groups(all P>0.05).Compared to the model group,50%PWTs of PTX+VitD_(3)group had significantly higher at d8,d12,d16 and d20(all P<0.05)after administration,TWL of PTX+VitD_(3)group had significantly higher at d4,d8,d12,d16 and d20(all P<0.001);serum 25-(OH)D_(3)levels of PTX+VitD_(3)group were significantly higher at d12 and d20(allP<0.001).Conclusions Intraperitoneal injection of vitamin D_(3)can alleviate paclitaxel-induced peripheral neuropathic pain in rats to a certain extent,providing a new direction of exploration for the management of clinical chemotherapy pain.
