摘要
采用响应面法优化虎奶菇菌丝体多糖的提取工艺,并对提取的多糖进行结构鉴定和抗氧化活性分析。结果表明,最佳提取条件为超声功率247.5 W、超声时间45 min、料液比1:36(g/mL),多糖提取率为(23.24±0.34)%,是水提法的2.2倍,优化工艺显著提高了多糖提取率。结构鉴定表明,提取的多糖基本不含核酸、蛋白质、色素等杂质,是一种含羧基和吡喃环的α型多糖,且没有三螺旋结构。抗氧化活性试验表明,虎奶菇多糖对ABTS自由基、羟自由基、超氧阴离子自由基、DPPH自由基均有清除能力,表现出较强的抗氧化活性。
Response surface methodology was used to optimize the extraction process of polysaccharide from the mycelium of Pleurotus tuber-regium,and the structure and antioxidant activity of the extracted polysaccharide were identified.The results showed that the optimal extraction conditions were as follows:ultrasonic power 247.5 W,ultrasonic time 45 min,solid-liquid ratio 1:36(g/mL),and the extraction rate of polysaccharide was(23.24±0.34)%,which was 2.2 times that of water extraction.This optimized process significantly improved the extraction rate of polysaccharide.The structural identification showed that the extracted polysaccharide basically contained no impurities such as nucleic acid,protein and pigment,and was anα-type polysaccharide containing carboxyl group and pyran ring,and had no triple helix structure.The antioxidant activity test showed that the polysaccharide had the ability to scavenge free radicals including ABTS,hydroxyl,superoxide anion and DPPH,showing strong antioxidant activity.
