摘要
本文研究了液相色谱法测定婴幼儿配方食品中乳铁蛋白含量的测定方法。样品加水溶解后,分别用磷酸调节pH等5种方法来沉淀蛋白,通过肝素亲和柱净化,紫外检测器检测。根据保留时间定性,外标法准确定量,检测计算回收率及精密度。结果表明,相比于其他方法,采用磷酸调节pH法沉淀蛋白方法的准确度和稳定性最好,同时该方法也适用于豆基乳粉和米粉样品中乳铁蛋白的检测。
The determination of lactoferrin content in infant formula food by HPLC was investigated.After the samples were dissolved with water,the protein was precipitated by five methods such as pH adjusted by phosphoric acid,purified by heparin affinity column and detected by UV detector,respectively.According to the retention time,the external standard method was accurate and qualitative,and the recovery rate and precision were detected and calculated.The results revealed that compared with other methods,the phosphoric acid regulated pH method was the most accurate and stable method for protein precipitation,and this method was also suitable for the detection of lactoferrin in soybean based milk powder and rice flour samples.
