摘要
目的研究法西多曲对大鼠心肌缺血再灌注损伤的保护作用。方法40只SD大鼠随机分为4组,分别为正常组(NC)、模型组(IR)、法西多曲低剂量(2.5mg/kg)组(FAS-L)和法西多曲高剂量(25mg/kg)组(FAS-H),每组10只。采用冠状动脉结扎法建立大鼠心肌缺血再灌注损伤模型,记录心率(HR)、左心室收缩压(LVSP)、左室舒张末压(LVEDP)、血管紧张素Ⅱ(AngⅡ)活性、AngⅡ受体(AT1)、血管紧张素转化酶(ACE)等指标变化。结果与NC组比,IR组HR减慢(P<0.01),LVSP显著降低,LVEDP明显增加(P<0.01),血浆及心肌中AngⅡ水平显著升高(P<0.01),心肌AT1和ACE免疫阳性表达明显增强。与IR组相比,FAS组HR、LVSP显著提高(P<0.05),LVEDP、AngⅡ显著降低(P<0.05),AT1和ACE免疫阳性表达明显减弱,且FAS-H组比FAS-L组更明显。结论法西多曲对心肌缺血再灌注损伤的保护作用可能与减少循环及心肌局部AngⅡ生成有关。
Objective To study the protective effect of facidor on myocardial ischemia-reperfusion injury in rats.Methods Forty SD rats were randomly divided into 4 groups:normal group(NC),model group(myocardial ischemia reperfusion injury group,IR),low-dose facidotril group(FAS-L,2.5mg/kg),and high-dose facidotril group(FAS-H,25 mg/kg),10 in each group.Coronary artery ligation was used to establish a rat myocardial ischemia-reperfusion injury model.Changes of indicators such as heart rate(HR),left ventricular systolic pressure(LVSP),left ventricular end-diastolic pressure(LVEDP),angiotensinⅡ(AngⅡ)activity,AngⅡreceptor(AT1),angiotensin-converting enzyme(ACE),etc.were recorded.Results Compared with the NC group,the HR in the IR group was slowed down(P<0.01),the LVSP was significantly decreased,the LVEDP was significantly increased(P<0.01),the level of AngⅡin plasma and myocardium was significantly increased(P<0.01),and the myocardial AT1 and ACE were immunopositive.expression was significantly enhanced.Compared with the IR group,the HR and LVSP of the FAS group were significantly increased(P<0.05),the LVEDP and AngⅡwere significantly decreased(P<0.05),and the immunopositive expressions of AT1 and ACE were significantly weakened.Conclusion The protective effect of facidol on myocardial ischemia-reperfusion injury may be related to the reduction of circulation and the production of AngⅡin local myocardium.
